Cefaclor is one of the most important and widely used antibiotics particularly for children.
This antibiotic as reconstituted with water must be stored at 4oC for a period not more than 14 days. The storage temperature is very important to ensure the stability of the active ingredient of this drug, since the degradation will lead to, increase the bacterial resistance, recurrent infections, and spread the community with highly resistant bacteria. The stability of five Cefaclor oral suspensions in the Jordanian market was studied during the real storage period (14 days) while keeping them at 4oC.
High Pressure Liquid Chromatographic (HPLC) procedure was used which gave us a clear idea about the concentration of the active ingredient during the 14 days after reconstitution.
It was found that Cefaclor was still stable during this period and within the pharmacopoeias specifications.
The antimicrobial effect of Cefaclor on certain bacteria was experimented also, and found that its activity didn't change during this storage period. The effect of 25oC and 37oC temperatures, on degrading and decreasing the activity of this antibiotic were studied, as Cefaclor is one of ß -lactam antibiotics which have a very sensitive group and ring for temperature; hydrolysis of this ring leads to loosing drug activity against bacteria. At 25 oC It was found that Cefaclor lost 8- 18% of its activity on the 7th day , and 11-27% on the 14th day; this degradation rate differed form one pharmaceutical company to the other; the antimicrobial activity started to decrease on the 7th day. During storage of these suspensions at 37oC, as the case in Aqaba Gulf and Jordan Valley during summer days, the degradation rate started from the second day, reached 18-50% on the 7th day, and 42-72% on the 14th day, the antimicrobial activity started to decrease also on the second day.
