Using cyclodextrin for capillary electrophoresis in separations of amino acids

Tryptophan is an important and frequently used starting material in the chemical synthesis of a range of pharmaceuticals.

Tryptophan possesses a chiral centre and is commercially available in the D, L or racemic forms.

There is a strong requirement to obtain data on the optical purity of input batches of tryptophan raw material prior to its use in synthesis.

Cyclodextrins (CDs) and their derivatives are one of the most commonly use additives for chiral separations in CE.

Buffer additives were carried out in order to find the best electrophoretic conditions to separate D and L-tryptophan enantiomers amino acid using cyclodextrin as a chiral active component in the background electrolyte.

In this work, simple and fast capillary electrophoresis (CE) procedure with UV detection (at 214 nm) for chiral separation of amino acid using α, β, γ Cyclodextrins (CDs) was developed and optimized.

Through increasing the concentration of α-CD, better separation was achieved.
