A number of different genes are believed to be associated with human obesity.

β3-Adrenergic receptors, which are mainly expressed in white and brown adipocytes, control lipolysis and heat production induced by noradrenaline .

A polymorphism resulting in a switch from tryptophan to arginine at the 64th residue in the β3-adrenergic receptor gene, known as the ADRB3 Trp64Arg, was identified in 1995.

Researcher until now has used the PCR-RFLP technique to analyze the ADRB3 genotype of the patients (Trp/Trp, Trp/Arg or Arg/Arg), the reference protocol due to 1995.

This protocol is effective but it has some disadvantages, it gives on the agarose gel 4-6 small DNA bands, which are close in size to each other and cannot be clearly separated and counted, thus the interpretation of the electrophoresis pictures is not easy and may lead sometimes to false results.

In this study new primers were designed and a new PCRRFLP method was optimized and tested on 45 persons (15 of each possible SNP genotype) the results were compared with that of the reference protocol.

The new protocol leaded to 1-3 bands, and the difference of the bands sizes was increased, so they could be better separated and counted.

The new method provides an easy and more effective and accurate diagnostic tool for the interested researcher.
