Single Nucleotide Polymorphisms (SNPs) are becoming favored genetic markers.

A large variety of the techniques have been developed for screening this type of mutations.

But mostly, these techniques are expensive and require specialized equipments.

In our study, we worked on evaluating ARMS-PCR (Amplification Refractory Mutation System-Polymerase Chain Reaction) method.

The optimization process was done using genomic DNA samples from rheumatoid arthritis patients to detect the genotype STAT4 rs7574865 (G/T).

The study included 13 DNA samples, whose STAT4 rs7574865 genotypes were already known using DNA sequencing method.

The optimization was done using different primers concentrations and annealing temperatures.

The optimized ARMS-PCR reaction was applied to the samples, but two of them showed a different genotype from the genotype shown by sequencing reaction.

Our study has set the thermal conditions and primer concentrations for the ARMS-PCR reaction, for the genotyping STAT4 rs7574865 polymorphism, but it still needs some modifications to attain more specificity.

ARMS-PCR is an easy, applicable and not costly method.
