English Abstract

The study was carried out at Biotechneology Center / Agricultural Researchers Directorate /Ministry of Science and Technology laboratories during the period of 1/8/ 2017 – 12/5/2018 using tissue culture and mutation technique to evaluate the response of regenerated plants produced from stressed callus of potato variety Arizona class Elite.

Potato tubers were irradiated with six doses of Gamma radiation namely (0.0, 2.5, 5, 10, 15 or 20 Gry) with Cobalt 60 source (CO60) at Protection of radiations Center /Ministry of Science and Technology.

The In Vitro experiment included studying the effect of NaCl at levels (6, 8 or 10 ds.

m-1) on callus initiated from leaves of irradiated tubers contain.

Significant differences were determined from amongThe experiment was based on C.

R.

D, the mean values of treatments by employing the least significant differences (LSD) test, at 0.05 level.

The Results showed that: Gamma dose, 20 Gy gave highest rate in related to number of microtubers, Na+, Cl-, proline and carbohydrate (6.78 microtubers.

Shoots-1, 30.63 mg.

g-1, 19.06 mg.

g-1, 17.61 µg.

g-1, 3.96 mg.

g-1 ) respectively, while 15 Gy was superior in total yield of microtubers which reached 6.18 g.

Shoots-1.

The salt level 6 ds.

m-1 gave highest rate in callus fresh weight, number and yield of microtubers were recorded (208.8mg, 3.89 microyubers and 3.34 g ) respectively, but salt level 10 ds.

m-1 gave highest rate in callus contain, Na+, Cl- and proline reached (29.13 mg.

g-1, 29.76 mg.

g-1 and 20.99 µg.

g-1 ) respectively.

The interactions between 10 ds.

m-1 of salinity stress and 10 Gy dose of gamma radiation for Arizona recorded highest callus fresh weight reached ( 293.33 mg) in addition to highest number of microtubers for each 8 ds.

m-1 in 20 Gy dose (7.00), while highest yield was recorded at 6 ds.

m-1 with 15 Gy 12.91 g.
