Abstract EN
The current study was conducted in order to increase date palm Phoenix dactylifera L.
salt tolerance using plant tissue culture through the treatment of some additives used for freezing.
Date palm embryogenic callus resulting from apical bud using MS (Murashige and Skoog) medium was immersed and inoculated in solutions and media for conservation supplemented with sucrose at a concentration of (342.3 g/L) for seven days and at 27 °C in the dark and the fetal call in liquid nitrogen at -196 C for 6 and 12 A week, then the callus was grown on medium included, sodium chloride concentrations of 0, 1500, 3000 and 4500 mg/l.
The effect of the above factors of some its chemical components including protein, amino acid, proline, abyssic acid, sodium ions and chlorine.
The results showed that the effect of sodium chloride was significant.
The treatment of NaCl significantly increased the concentration of proline especially at concentration of 1500 ppm reached 60.30mg/g.
For the ionic content of fetal calcification, the effect of sodium chloride on the sodium ion concentration of the embryonic calf after conservation for 12 weeks was significant.
The treatment of sodium chloride at the third concentration was 4500 ppm.
The highest concentration of sodium ions was 33.7 mg/g dry weight.
The sodium chloride treatment was superior at a concentration of 4500 ppm, the highest concentration was 15.98 mg.
g-1 dry weight.
