Acacia nodules were collected from El Daein-Sudan and washed thoroughly under running tap water then dried and subjected to grinding with the aid of a mechanical grinder to form powder, sieved and stored in a dry container.10g of acacia powder was dissolved in 100ml of absolute alcohol to give a concentration of 100mg plant product/ml of absolute alcohol.
then sodium chloride 10 gand glacial acetic acids 10 ml were added.
another 10g of acacia powder was dissolved in 100ml of ionized water to give a concentration of 100mg plant product/ml of water.
then sodium chloride 10gand glacial acetic acid 10 ml was added.
the tissue fixed with acacia alcoholic solution was excellently stained by hematoxylin and eosin the nucleus stained bluish with ping or magenta background as same the tissue fixed in formalin but, the tissue materials fixed in ionized water acacia mixture failed to color the tissue elements.
