الهدف من هذه الدراسة هو دراسة كيفية حدوث الإسهال في الحيوانات المصابة بطفيل الكريبتوسبوريديوم حتى يمكن تمهيد الطريق لوضع استراتيجية علاجية فعالة و مؤثرة.
تمت الدراسة على نوعين من الحيوانات (العجول و الفئران).
تم فحص عينات من البراز للعجول المصابة (العدد 12)، و ذلك للحصول على أطوار الطفيل (Oocysts & sporozoites)، كما تم الحصول على عينات البراز التي تحتوي على مجموعة التفاعل التي تحدث في حالات الإسهال و كذلك تم استخدام Oocysts في إحداث الإصابة في الفئران.
كما تم استخدام الــ sporozoites و عينات البراز في حجرة يوسينج (Ussing chamber) لمعرفة تأثير كل منهما على معدل التغيير في أيونات الصوديوم و أثرهما في إمكانية حدوث الإسهال، مقارنة بعينات مماثلة من العجول الغير مصابة (العدد 6).
و بعد إحداث الإصابة في الفئران (العدد 24)، تم أخذ عينات من الأمعاء المصابة لــ 12 فأر للحصول على المواد التي تفرز منها كرد فعل للإصابة، و تم دراسة تأثيرها في إحداث الإسهال و ذلك في حجرة يوسينغ، مقارنة بعينات من أمعاء الفئران الغير مصابة (العدد 6)، كما تم دراسة باثولوجيه للأمعاء المصابة لعدد 12 فأرا مصابا.
ثم تم إصابة مجموعة أخرى من الفئران (العدد 6)، و ذلك لعمل نموذج بوضع العلاقة بين Oocysts و مدة الإصابة بالأيام.
اتضح من البحث أن عينات البراز للعجول المصابة و أمعاء الفئران المصابة و الأسبوروزريت Sporozoites بها مواد سامة لها دور أساسي في آلية حدوث الإسهال و أن هذه المواد تتأثر بالجرعة و الوقت و الحرارة، و بذلك يمكن معالجة هذه الإصابة بإعطاء مضادات لهذه المواد السامة.
و عند فحص عينات الأمعاء للفئران المصابة باثولوجيا، وجد أنه يوجد درجات متفاوتة من الالتهاب في خلايا الأمعاء و أن عددا كبيرا من الخملات (intestinal villi) قد دمر في المعي الصائم (jejunum) و المعي اللفائفي (ileum) في اليوم الخامس بعد الإصابة، هذا بالرغم من أن عينات البراز من هذه الفئران كان شبه متماسك (semiformed).
و عند عمل نموذج لإفراز الأووسيست oocysts في الفئران المصابة، وجد أنه يبدأ في اليوم الثالث من تناول الجرعة، ثم يصل لأعلى نسبة في اليوم التاسع، و يختفي بعد أسبوعين.
و أثبت البحث فعالية استخدام الفئران كحيوانات تجارب رخيصة بدلا من إحداث الإصابة في العجول.
Abstract EN
The present work was planned to study the possible mechanism(s) of diarrhea in cryptosporidiosis.
Stool eluates and intestinal homogenates from naturally and experimentally infected animals together with purified sporozoites were examined utilizing Ussing chamber to demonstrate their possible enterotoxic effects.
Besides, the histopathological changes in experimentally infected mice were studied to assess the possible underlying mechanism(s) of diarrhea in cryptosporidiosis.
Characterization of the enterotoxic activity associated with cryptosporidiosis may have a great impact on the development of effective strategies for its treatment.
The results of the present study revealed that in intestinal cryptosporidiosis, enterotoxic substances are secreted which likely induce diarrhea.
They induce an increase in the transepithelial potential difference (Disc) to reach its maximum after IS minutes and then slowly decrease to reach the baseline after 55 minutes (for stool eluates) and 35 minutes (for intestinal homogenates).
On the other hand, the purified sporozoites showed an increase in Disc after 9 minutes and then a decrease after that to become maintained at relatively high level.
The enterotoxins were found to be time and dose-dependent and heat-labile.
The osmotic gap showed that the mechanism of diarrhea is rather secretory.
In experimentally infected mice, shedding of oocysts first appeared iri stools-.
3 days postinoculation (PI), reaching a peak 9 days (PI) and disappeared 15 days PI.
Although the infected mice showed mild to severe degree of intestinal inflammation (marked villous atrophy and crypts hyperplasia), the stool was semiformed.
Besides, the present study showed marked validity of mouse as an in vivo model to study the pathophysiology of cryptosporidial diarrhea.
This model is inexpensive and serves as a suitable alternative to neonatal calves for efficient oocyst propagation.
