تم إجراء هذا البحث لمقارنة التراكيب الوراثية بين النخيل المكاثر بواسطة الطرق التقليدية (الفسائل) و النخيل المكاثر بواسطة زراعة الأنسجة باستخدام تقنية RAPD.
استخدم في هذه الدراسة أس أكثر من 80 بادئ.
أعطت ثمانية منها اختلاف في عدد و نوعية الحزم (Bands) الناتجة من تضخيم (Amplification) ال د.
ن.
أ (DNA) لكل من النخيل النسيجي و الفسيلي.
نتائج هذا البحث تبين أن تقنية RAPD يمكن أن تستخدم للتحقق من مدى مطابقة التراكيب الوراثية للنباتات المكثرة بواسطة زراعة الأنسجة عن طريق تكوين الأجنة الخضرية مقارنة بالأصل الفسيلي.
و قد نوقشت في هذا البحث الأسباب المحتملة للاختلافات التي ظهرت بين التراكيب الوراثية (عدد و نوعية الحزم) للنخيل المكثر بزراعة الأنسجة و أصوله الفسيلية.
Abstract EN
RAPD techniques were used to compare the tissue-culture produced date palm trees with their offshoot origins.
Eight random primers were successfully used to amplify DNA from OSO (Off-Shoot Origin) and TCP (Tissue Culture Produced) date trees and gave sufficient polymorphism for each cultivar.
Our result shows that RAPD (Random Amplified Polymorphic DNA) technique can be successfully applied to determine the genetic fidelity of micro propagated plants of date palms.
Possible explanations for the variations detected in micro propagated date palm via callus and embryogenesis were discussed.
