للجذور لحرة و أكسدة الدهون دور في الأضرار ببروتينات الخلية و كذلك الحامض النووي الديؤكسي ريبوزي و الدهون.
و يوجد بالخلية وسائل دفاعية ضد أضرار هذه الجذور مثل إنزيم السوبر أوكسيد ديسميوتيز و إنزيم الجلوتأثيون بير أوكسيديز و مادة الجلوتأثيون المختزلة.
و قد أثبتت الدراسات أن لخلايا المسرطنة لها القدرة على إنتاج الجذور الحرة أكثر من الخلايا الطبيعية و ذلك نتيجة للنمو السريع للسرطان مما يؤدي إلى نقص الجلوكوز و لأوكسجين.
كما أثبت أيضا أن الجذور الحرة لها دور فعال في تثبيط الموت المبرمج للخلية لذلك تم استخدامها كجزء من العلاج الكيميائي للقضاء على الخلايا السرطانية سريعة النمو دون الخلايا الطبيعية.
شملت هذه الدراسة مجموعة من النساء المصابات بسرطان الثدي و مجموعة أخرى من النساء المصابات بـأمراض حميدة في الثدي.
و تم اخذ عينات من النسيج السرطاني و كذلك عينات من نسيج الثدي السليم باثولجيا و قياس نواتج أكسدة الدهون و مضادات الأكسدة.
بالإضافة إلى قياس نسبة الحامض النووي (د.
ن.
أ) و نشاط إنزيم (كاسبيس -3) و أكسيد النيتريت.
و قد أظفرت نتائج هذه الدراسة عن ارتفاع نسبة نواتج أكسدة الدهون، أكسيد النيترتيت و مضادات الأكسدة و نسبة نشاط إنزيم كاسبيس-3 و الحامض النووي (د.
ن.
أ) في الأنسجة السرطانية عن الأنسجة الحميدة و قد تبين أن استخدام نسبة نواتج أكسدة الدهون، أكسيد النيتريت و مضادات الأكسدة و نشاط إنزيم كاسبيس-3 و الحامض النووي (د.
ن.
أ) يزيد من حساسية اكتشاف المرض بنسبة98 %-% 84-% 74-86 %-90 %-94 % على التوالي و بخصوصية 93.3 %-86.7 %-66.7 %-% 93.3-87 % على التوالي و بلغت 100 % في الحامض النووي (د.ن.
أ).
و بين النسبة بين نواتج أكسدة الدهون على مضادات الأكسدة، و بين أكسيد النيترتيت و نشاط إنزيم كاسبيس-3 و الحامض النووي (د.
ن.
أ).
و من ذلك نستنتج ارتفاع نسبة الأكسدة و أكسيد النيترتيت و كذلك الموت المبرمج الفسيولوجي في سرطان الثدي خاصة في المراحل المتأخرة من الورم.
Abstract EN
Reactive oxygen species are important in the pathogenesis of many diseases, including breast cancer.
In this study, we aimed to evaluate oxidative stress in patients with breast cancer and to investigate its relationship with apoptosis.
Our results showed that, the median levels and positivity rates of Malondialdehyde (MDA), Nitric Oxide (NO), Total Antioxidant (TAO), caspase-3, % DNA fragmentation and MDA / TAO ratio measured in breast cancer tissues by colorimetric methods were higher in the malignant group as compared to benign control group.
Moreover, MDA, NO and % DNA fragmentations were over produced in advanced grade and stage P < 0.05.
Furthermore, a significant positive correlation was found between MDA / TAO ratio and positive lymph node metastasis.
Also, there were significant positive correlations of caspase-3 and %DNA fragmentation with positive estrogen receptor and NO.
Moreover, the total antioxidants were positively correlated with positive progesterone receptor.
In Conclusion; oxidative stress, NO and apoptosis are highly detected in breast cancer tissues especially with advanced grade and stage.
