تم إجراء هذه الدراسة لتقييم استجابة المناعة الخلوية و الدموية في المرضى المشتبه في إصابتهم بطفيل التوكسوبلاما و قد تم استخدام سلالة التكسوبلازما و تم استخدام فئران التجارب 400 فأر بمعدل 10 فئران في كل مرة للحفاظ على سلالة التكسوبلازما بالتمرير بانتظام و كذلك بغرض تحضير الأنتيجين لاستخدامه في إجراء الاختبارات. أجريت هذه الدراسة على 76 شخص منهم 56 مريض مشتبه في إصابته بمرض التوكسوبلازموسز حيث تم تقسيم المرضى إلى أربعة مجموعات (أ) 17 مريض يشتكون من أعراض بالعين تشير إلى الإصابة بمرض التوكسوبلازموسز، (ب) 23 مريض لهم مشاكل خاصة بأمراض النساء و التوليد، (ج) تشمل 9 أطفال مشتبه في إصابتهم بمرض التوكسوبلازموسز الوراثي، (د) 7 مرضى مصابون بتضخم الغدد الليمفاوية نتيجة الإصابة بمرض التوكسوبلازموسز بالإضافة إلى 20 شخص سليم كمجموعة ضابطة يتمتعون بصحة جيدة و لا يشكون من أي أعراض لمرض التوكسوبلازموسز. و قد تم إجراء الاختبارات الآتية : (1) اختبار اليزا لتعيين الأجسام المضادة جي و أم (2) اختبار تحول الخلايا الليمفاوية لتعيين رد فعل التفاعل النوعي للخلايا الليمفاوية و قد تم قياس رد فعل التفاعل النوعي للخلايا الليمفاوية. و قد أظهرت النتائج أن من بين المجموع الكلي لعدد 56 مريض، 49 مريض (87,5%) أعطوا نتائج إيجابية ذوي تفاعل مصلي إيجابي باستخدام الأنتيجين المحضر من التسكوبلازما في واحد أو أكثر من الاختبارين المستخدمون في الدراسة و أن عدد 7 من المرضى أعطوا نتائج سلبية.
و قد تم التوصية بمتابعتهم لاستبعاد إصابتهم بطفيل التكسولازما. و قد توصلت الدراسة إلى ارتفاع معدل إيجابية الأجسام المضادة لطفيل التكسوبلازما بين المرضى المتوقع إصابتهم مما يدل على كفاءة اختبار اليزا في تشخيص الإصابة بطفيل التكسوبلازما و أن التفاعل المناعي للخلايا الليمفاوية يؤكد تلك النتائج و أعلن عن إصابة كامنة في بعض الحالات و يمكن استخدام تلك القياسات في العلاج.
Abstract EN
Objective : the study was conducted to assess the hum oral and cellular immune responses in patients suspected to have Toxoplasma infection, and to evaluate their use as diagnostic tools..
Subjects and Methods : Seventy six persons were enrolled in the study, 56 of them are patients with toxoplasmosis.
In addition to 2o apparently healthy persons were served as control.
They were divided into 4 main groups according to their signs and symptoms, Group I, 17 patients with ocular signs and symptoms suggestive of toxoplasmosis Group II, 23 patients complaining of complicated obstetric problems, Group III, 9 infants and children with signs and symptoms of congenital toxoplasmosis, and Group IV, 7 patients with manifestations suggesting toxoplasma lymphadenopathy.
RH Toxoplasma strain was used.
Experimental animals: 400 mice were used, 10 each time, for maintenances of Toxoplasma strain, by regular serial passage and for preparation of antigen.
Enzyme linked immunosorbent assay (ELISA) for detection of anti-Toxoplasma IgG and IgM antibodies.
Lymphocyte transformation test were done for detection of specific lymphocyte proliferation (blast genic response).
Results: out of total 56 patients, 49 patients (87.5%) gave positive results to Toxoplasma antigen in one or more of the specific techniques applied.
Seven patients gave negative results were recommended for periodic follow up before exclusion of toxoplasmosis.
Conclusion : high rates of seropositivity detected among suspected patients suggest the reliability of ELISA in diagnosis of toxoplasmosis.
Specific lymphoblastognic response would confirm the results and reveal dormant infection in some suspected subjects.
However, rising titer/ seroconversion is a prerequisite for therapy.
