يهدف البحث إلى دراسات كيميائية و تقييم الفاعليات البيولوجية لبعض النباتات البرية المنتقاه من العائلة الشفوية و هم نبات الزعتر و نبات الزعيتران و نبات المردقوش حيث أنها تحتوي على مواد فعالة كمضادات للأكسدة.
و يشمل البحث تحليل مكونات الزيوت الطيارة للنباتات باستخدام كروموتوجرافيا لاالغاز المدمج بطيف الكتلة و قد أسفر عن وجود عدد من المركبات تم التعرف عليها و التي تمثل حوالي 95 % من مكونات الزيوت الطيارة للنباتات.
و تضمنت الدراسة البيولوجية دراسة التأثير المضاد للأكسدة للزيوت الطيارة لكل من النباتات باستخدام TEAC assay, DPPH assay و مقارنة النتائج بمادة الترولكس كمادة مرجعية.
كما تضمنت الدراسة تأثير الزيوت الطيارة للنباتات على مدى تسمم خلايا الكبد المزروعة لفئران التجارب و مدى حمايتها لخلايا الكبد باستخدام تركيزات مختلفة لكلا منها مع مقارنتها بمركب السليمارين كمادة مرجعية.
و قد استنتجت الدراسة إلى وجود دلائل إيجابية للزيوت الطيارة لنباتات الزعتر و نبات المردقوش كمضادات للأكسدة و كوقاية للكبد.
Abstract EN
Objectives : to isolate the essential oils of Origanum syriacum, Thymus decussates and Salvia multi caulis and evaluate their ant oxidative and hepatoprotective activities against paracetamol induced injury in primary-cultured rat liver cells.
Methods : the essential oils isolated from Origanum syriacum, Thymus decussates and Salvia multi caulis were analyzed by GC-MS.
Antioxidant activities of the essential oils were determined by DPPH scavenging activity (DPPH) and Trolox equivalent antioxidant capacity (TEAC) asssays.
Conditions have been investigated to devise an in-vitro colorimetric MTT assay method for the hepatoprotective activity against paracetamol induced injury in primary-cultured rat liver cells.
Results : the major component of the essential oils of Origanum syriacum.
Thymus decussates was thyme 58.6% and 21.6 % respectively.
While the major component of Salvia multi caulis was camphor 39.8 %.
The antioxidant activity showed that the volatile oil of Origanum exhibited the highest activity followed by Thymus then Salvia.
As for the hepatoprotective activity, the assay revealed that the volatile oil of Salvis multi caulis showed a suitable safety margin (the toxic concentration was 125 ug / m, while hepatoprotective concentration was only 25ug / ml).
Conclusion : the essential oils of Origanum syriacum, Thymus decussates and Salvia multi caulis exhibited remarkable antioxidant activity and significant hepatoprotective activity.
