الهدف من الدراسة : دراسة الدور الذي يلعبه الإجهاد التأكسدي في إحداث الأثر السام لعقار السيسبلاين و إذا كان هذا صحيحا فهل يعد استخدام مضادات الأكسدة فعالا في الحد من سمية هذا العقار.
تحديد بعض آليات الحماية الممكنة بواسطة استخدام الكارنتين أو حمض الليبويك.
طرق البحث : في هذا البحث، تم دراسة تأثير الإعطاء المترافق لمادة الكارنتين (500 مج / كج) و لمضاد الأكسدة الطبيعي و كاسح الشقوق الحرة حامض الليبويك (100 مج / كج) كلا على حدا على التأثير السمي لعاقر السيسبلاتين (10 مج / كج) على أنسجة القلب في الجرذان و ذلك بقياس المؤشرات الدالة على سلامة عضلة القلب و الإجهاد المؤكسد في المجموعات المختلفة و مقارنتها بكلا من المجموعة الضابطة و تلك التي حقنت بالسيسبلاتين.
لذا فقد تم قياس مستويات إنزيمات القلب في مصل الدم و تركيز التروبونين في البلازما كدلالة على سلامة خلايا القلب، بالإضافة إلى فوق أكسدة الشحوم في أنسجة القلب و محتوى مادة الجلوتاثيون و مستوى نشاط إنزيم السوبر أوكسيد ديسميوتيز علاوة على محتوى البروتين في أنسجة القلب.
نتائج البحث : لقد ثبت بصورة جلية تأثير عقار السيسبلاتين السام على القلب متمثلا في الارتفاع الملحوظ لمستويات إنزيمات القلب في مصل الدم و تركيز مادة التروبونين في البلازما بالمقارنة بالمجموعة الضابطة.
و قد ترافق هذا الارتفاع مع زيادة ملحوظة في فوق أكسدة شحوم أنسجة القلب و استنفاد مادة الجلوتاثيون و انخفاض مستوى نشاط إنزيم السوبر أوكسيد ديسميوتيز مما يشير إلى الإجهاد المؤكسد الذي أحدثه عقار السيسبلاتين في أنسجة القلب و بالتحليل الإحصائي للنتائج فلقد لوحظ أن الكارنتين أو حمض الليبويك قد أضعف بشكل كبير الخلل الذي أحدثه عقار السيسبلاتين في المؤشرات المذكورة أعلاه.
الاستنتاج : الإعطاء المترافق للكارنتين و حمض الليبويك و كان له أثر واق ضد الإجهاد المؤكسد المصاحب لتعاطي عقار السيبلاتين في الجرذان.
Abstract EN
Objectives : Increased oxidative stress has been implicated in the cardio toxicity that limits the clinical use of cisplatin as an anti-tumoral drug.
Therefore, this study was conducted to evaluate the cytoprotective potential of acetyl-L-carnitine, DL-a-lipoid acid against cisplatin-induced myocardial injury in male rats.
Materials and Methods : Sixty male albino rats were divided into six groups.
The first three groups were treated with normal saline, acetyl-L-carnitine (500 mg / kg, i.
p.) and DL-a-lipoid acid (100 mg / kg, p.
o.) respectively, for 10 successive days.
The 4th, 5th and 6th were treated with the same doses of normal saline, acetyl-L-carnitine and DL-a-lipoic acid, respectively, for 5 successive days before and after a single dose of cisplatin (10 mg / kg, i.
p.).
Serum LDH, CK, CK-MB and plasma cTnl concentration were estimated.
In addition, malondialdehyde (MDA), glutathione (GSH) contents, superoxide dismutase activity (SOD) and protein content in cardiac tissues were measured.
Results : Cisplatin-treated rats experienced a significant elevation of serum activities of LDH, CK, CK-MB and cTnl plasma concentration.
These effects were accompanied by a significant increase in MDA level and a significant decrease in GSH content, SOD activity and total protein content.
However, acetyl-L-carnitine and DL-a-lipoid acid significantly attenuated the cisplatin-evoked disturbances in the above-mentioned parameters.
Conclusion : Acetyl-L-carnitine and DL-a-lipoid acid were proven to be potential candidates to ameliorate cardio toxicity associated with cisplatin in rats.
