ثلاث عترات من البكتيريا اللاهوائية المحبة للحرارة و للملوحة المتوسطة الموسومة ب HAR100T HAR720T HAR200T تم عزلها من موقعين للينابيع الساخنة في الأردن .و تتصف هذه العترات بأنها تنمو في وسط ملحي 12-15% من كلوريد الصوديوم و لها القدرة على النمو لاهوائيا (و ليس هوائيا) على درجات حرارة 37-63 مئوية ؛ و هي عصيات قصيرة متحركة و متجرثمة و تفاعلها إيجابي مع صبغة غرام.

وجدت هذه الدراسة أن محتوى قواعد (الجوانين + السيتوسين) في الحامض النووي لهذه العترات يتراوح ما بين 43-54% أما أن تواجد أو غياب إنزيمات الاميليز و البروتييز و الفوسفاتيز القلوية و إنزيمي جلوكوز 6 فوسفات و 6 فوسفو جلوكونيت النازعين للهيدروجين و محتواها من الدهنيات القطبية و مقاومتها لبعض الأدوية و البصمة الوراثية لمحتوى دي .ان.

اي.

العشوائي و تحليل بصمة sDNA 16 تثبت أن هذه العترات الثلاثة تختلف اختلافا جذريا بالصفات الجينية و النوعية مما يدعم كونها عترات جديدة من الجنس .

Paenibacillus

Abstract EN

Three strains of facultative anaerobic thermo tolerant and moderately halophile bacteria were isolated from two Jordanian hot spring locations.

The novel Bacillus strains were short rods, Gram-positive, motile and spore formers.

The strains were capable of anaerobic (but not aerobic) growth at 63oC, moderately halophile, able to grow at NaCl concentration range of 12-15 % and at temperatures range of 37-63oC.

Moreover, the strains were resistant to aztreonam, bacitracin and oxacillin.

The range of the guanine-plus-cytosine content of the strains DNAs was 43–54 mol %.

The three strains HAR7100T.

HAR720T and HAR200T, differ phenotypically from each other in terms of antibiotic resistance, compositions of polar lipids, presence and absence of several enzyme activities such as amylase, protease alkaline phosphatase, G6PDH and 6PGDH.

Moreover, the novel Bacillus strains were shown to differ genotypic ally by random amplified polymorphic DNA fingerprinting and 16S r DNA amplification patterns.

Phenotypic and genotypic variations may support establishment of new strains of the same genus Paenibacillus.
