Ten species belonging to genus Aphis were collected from a different locality of Egypt.
Genetic polymorphisms in these species were investigated by isozymes, SDS- PAGE and RAPD-PCR.
Four isozymes (a-and P-esterase, acid phosphatase, and alkaline phosphatase) revealed high level of polymorphism among the studied species.
RAPD-PCR was carried out using ten primers.
The primer C15 was not effective in revealing polymorphism among the studied species (0% polymorphism).
Amplification products of the ten species produced 88 reproducible bands, from which 76 (87.4%) were polymorphic.
The results generated from isozymes, SDS-protein and RAPD-PCR profiles were used to draw the genetic relationships among the studied species.
The constructed dendrogram tree divided the ten species into two main groups, the first group divided into two subgroups; the first included A.
punicae, whereas, the second contained A.
gossypii, A.
medicaginis, A.
fabae, A.
citricola, A.
compositae and A.
craccivora.
The second main group included A.
rumicis, A.
zizyphi and A.
nerii.
Twenty four out of 139 total protein, isozymes and RAPD markers were found to be species specific.
Nine of the ten Aphis species had species-specific markers.
On the other hand, eleven out of 16 markers showed band-specific markers, including a and P-esterase, acid and alkaline phosphatase isozymes, total protein and primers BIO, D5,117, L12, L13 andZl.
