يعد إلتهاب الضرع الإكلينكي مشكلة رئيسية و خطيرة في المزارع الحلابة بالرغم من الجهود التي تتخذ في التخلص من هذا المرض.
وقد أجريت هذه الدراسة لتقييم مدى إنتشار هذه المشكلة في 8 مزارع مختلفة الإسماعيلية و تم الفحص الإكلينيكي على عدد 3000 و 1500 من الأبقار و الجاموس الحلابة على التوالي.
و قد أخذت 130 عينة من 108 حيوانات حلابة تعاني من التهاب الضرع الإكلينيكي.
أجريت إختبارات بكتيرولوجيه و بيوكيميائية لفحص تلك العينات و تم عزل عدد 56 (53.1 %) و 34 (26.2 %) و 4 (3.1 %) من ميكروبات المكور العنقودي الجلدي و المكور العنقودي الذهبي و المكور السبحي يوبريزو انتيروباكترا جلوميرانس و الباسيلس انثراكويد على التوالي و بقية العينات (18.5 %) لم تظهر أي عزل بكتيري.
و تم أيضا إجرء تقنية تفاعل البلمرة المتسلسل كإختبار سريع و حساس و دقيق في تشخيص ميكروب المكور العنقودي الذهبي لأنه أشد ضراوة و مسبب لخسائر فادحة في مزارع إنتاج اللبن وتم فحص جميع عزلات هذا الميكروب من عينات اللبن لحالات التهاب الضرع الإكلينيكي باستخدام هذه التقنية باستخدام بادئات خاصة بهذا الميكروب أوضحت النتائج أن جميع (100 %) العزلات قد كانت إيجابية.
من هذه النتائج يمكن أن نستخلص أن تقنية تفاعل البلمرة المتسلسل يعتبر طريقة فعالة في تشخيص المرض حيث تم إجراؤه في مدة قصيرة.
Abstract EN
Commercial dairy farms had serious mastitis problems in spite of its control hygienic measures, for detection of this problem, a total of 3000 and 1500 lactating cows and buffaloes respectively were examined for clinical mastitis in 8 different farms at Ismailia governorate.
130 milk samples were taken from 108 dairy animals suffering from clinical mastitis.
the bacteriological examination revealed that 56 (43.1 %), 34 (26.2 %), 8 (6.1 %), 4 (3.1 %) and 4 (3.1 %) from clinically mastitis milk samples were positive for staphylococcus epidermis's, staphylococcus aurous, streptococcus uteri's, bacillus anthropoid and enterobacter agglomerans, respectively.
while 24 (18.5 %) of the samples were negative.
polymerase chain reaction (pcr) was also conducted to staphylococcus aurous isolates as a major pathogenic bacteria in cases of mast-it is, genomic dnas extracted from all s.
aurous isolates (n = 34) were successfully identified by this pcr assay.
pcr was proved to be specific and sensitive in diagnosis of clinical mastitis.
