هذه الدراسة تهدف لعزل ميكروب منهيميا هيموليتيكا من الأغنام المصابة بالتهابات رئوية و التأكد من عزله بواسطة إختبار البلمرة، و التعرف على أنواعه، بالإضافة إلى معرفة حساسية هذا الميكروب للمضادات الحيوية.
فقد تم عزل 52 (20.8) عزلة منهيما هيمولتيكا من 250 عينة (120 عينة رئوية، 90 مسحة أنفية و 40 عينة من اللوز) تم تجميعها من مجازر محلية و فحصت في هذه الدراسة.
من 52 عزلة من هذا الميكروب ب 42 (80.8 %) عزلة من النوع و (19.2 %) عزلة Tمن النوع.
كما تم إختيار أربع معزولات عشوائية من 52 عزلة للتأكد منها بواسطة اختبار البلمره وذلك للمقارنة بين إختبار البلمره و الإختبارات الكيميائية في الكشف عن ميكروب المنهيميا هيموليتيكا.
تم إختيار حساسية هذه المعزولات لعدد من المضادات الحيوية و كان جميع الميكروبات حساسه للدانوفلوكساسين و الأنروفلوكساسين، بالإضافة إلى معزولات كانت حساسة بنسب متفاوتة للجينتاميسين، الستربتوميسين 85.5 %، 67.9 %، 69.2 % على التوالي.
كما وجد أن جميع المعزولات كانت مقاومة الكلنداميسين، الأوكسيتتراسيكلين و السلفافيورازول.
باللإضافة إلى الأموكسيسيلين، و الأوكسيسيلين، الأمبيسلين، التتراسيكلين و السلفافيورازول.
بالإضافة إلى الأموكسيسيلين، الأوكسيسيلين، الأمبيسلين، التتراسيكلين، الأرثروميسين و البنسلين وذلك بنسب 67.3 %، 67.3 %، 59.6 %، 50 %، 34.6 %، 30.8 % على التوالي.
Abstract EN
This study aimed at investigating isolation, identification and confirmation of the isolated strains by biochemical tests and PCR, also biotyping of Mannheimia haemolytica strains isolated from sheep with pneumonia.
In addition, antibiotic susceptibility of the isolates were determined against a member of antimicrobial agents.
Mennheimia haemolytica was isolated in percentage of 52(20.8 %) from a total 250 samples, (120 pneumonic lungs, 90 Nasopharyngeal swabs, 40 Tonsils samples) were examined in the present study.
All samples were obtained from local state-run slaughterhouse.
A total of 52 Mannheimia haemolytica strains, were isolated and identified.
Out of 52 Mannheimia haemolytica strains, four random strains were chosen and confirmed by PCR and the result indicate that the PCR go in accordance with those of biochemical tests.
From 52 sheep isolates, 42 (80.8 %) and 10 (19.2 %) were biotyped as biotype A and biotype T, respectively.
Biotype A strains of Mnnheimia haemolytica isolated in this study is more frequently isolated from sheep lungs and utilized arabinose better than xylose and did not ferment trehalose.
Therefore, it may be suggested that fermentation of arabinose and trehalose are more reliable than that of xylose when performing biotyping.
In vitro antimicrobial susceptibility test results indicated that all Mannheimia haemolytica strains isolated were tested for their susceptibility to various antimicrobial agents using a disc diffusion method.
All 52 sheep isolates tested, were susceptible to enrofloxacin and danofloxacin.
In addition, isolates were found susceptible to gentamicin, streptomycin, and kanamycin with percentage 86.5 %, 76.9 %, and 69.2 % respectively.
All 52 isolates tested were resistant to clindamycin, oxytetracycline and sulfafurazole.
In addition, amoxicillin, oxacillin, ampicillin, tetracycline, erythromycin and penicillin G.
