كان الهدف من هذا العمل البحثي هو إثبات أن هناك علاقة بين وجود الفيروس الحليمي الآدمي و التحول السرطاني في أنسجة الرأس و الرقبة و علاقة ذلك بعملية التساقط الحيوي للخلايا من خلال إنتاج بروتين ال السي أي إيه بي – 2 مما يؤدي إلى تسرطن هذه الأنسجة. تضمن البحث (30) حالة من حالات سرطان الرأس و الرقبة حيث تم تقسيمهم إلى مجموعتين : • المجموعة الأولى : تشمل عينات من الأنسجة المصابة بسرطان الرأس و الرقبة. • المجموعة الثانية : تشمل عينات من الأنسجة الطبيعية المحيطة بالورم من نفس المرضى. تم تجميع عينات الأنسجة من جميع المشتركين في الدراسة و تم فحص الأنسجة خلويا, كما تم استخراج و قياس الحمض النووي دنا للفيروس الحليمي الآدمي باستخدام تفاعل البلمرة المتسلسل (PCR).
و تم أيضا استخراج و قياس الحمض الريبوزي النووي رنا لين ال CIAP-2 بطريقة تفاعل البلمرة المتسلسل العكسي النوعي و طريقة تفاعل البلمرة المتسلسل العكسي شبه الكمي. و قد أثبتت طريقة تفاعل البلمرة المتسلسل (PCR) وجود الفيروس الحليمي الآدمي في 86.7% من الحالات مما يثبت وجود علاقة قوية بين الإصابة بالفيروس و حدوث سرطان الرأس و الرقبة.
و أثبتت طريقتي تفاعل البلمرة المتسلسل العكسي النوعي و الشبه الكمي وجود جين ال CIAP-2 في 76.7% من الحالات مما يثبت تأثير الجين على حدوث سرطان الرأس و الرقبة.
بالإصابة وجود جين ال CIAP-2 في 73.1% من مرضى السرطان المصابين بالفيروس الحليمي الآدمي (عينات السرطان فقط) مما يؤكد وجود علاقة قوية بين وجود الفيروس و تأثيره على عملية التساقط الحيوي للخلايا من خلال إنتاج جين ال CIAP-2.
Abstract EN
It was relevant to assess the impact of a HPV-16 (human papillomavirus-16) which is a high-risk genotype on head and neck cancers and its relation to CIAP-2 expression (Cellular Inhibitor of apoptosis-2).
Tissue samples were taken from patients diagnosed as head and neck cancer.
All samples were subjected to qualitative polymerase chain reaction (PCR) analysis for HPV, qualitative and semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) of CIAP-2.
There was a significant association between HPV-16 and CIAP2 positivity and head and neck tumors.
CIAP-2 expression in malignant tissues was highly associated with HPV-16 infection at sensitivity 73.9 % and absolute specificity.
