أجري هذا البحث لدراسة تأثير إنوسيتل سداسي الفوسفات (IP6) على مناعة الجسم و قدرته على مقاومة الأمراض.
و لهذا الغرض شملت هذه الدراسة ثلاث مجموعات من ذكور الجرذان البيضاء.
المجموعة الأولى استخدمت كمجموعة ضابطة.
المجموعة الثانية حقنت ببكتريا إيروموناس هيدروفيلا مرة واحدة أسبوعيا لمدة أربعة أسابيع (0,2 ملي لتر–تركيز 2×610).
المجموعة الثالثة حقنت نفس الجرعة من بكتريا إيروموناي هيدروفيلا و في ذات الوقت أعطيت IP6 ثلاث مرات أسبوعيا لمدة أربعة أسابيع (بلغت الجرعة الكلية للـ IP6 40 ملي جرام / كيلو جرام من وزن الجرذ).
تم دراسة النشاط البلعمي لخلايا الدم البيضاء و كذلك مستوى كلا من بروتينات البلازما و الأجسام المضادة باستخدام عملية الترحيل الكهربي.
تم أيضا تقدي بعض الإنزيمات المضادة للأكسدة و مستوى الشق الحر لأكسيد النيتروز (NO).
و أكدت النتائج على زيادة النشاط البلعمي لخلايا الدم البيضاء و زيادة و مستوى الشق الحر لأكسيد النيتروز (NO) في المجموعة الثانية مع انخفاض إنزيمات hepatic catalase و total peroxidase و Super oxide dismutase (SOD).
على الجانب الآخر فأن إعطاء IP6 لجرذان المجموعة الثالثة قد حسن بوضوح من نشاط الإنزيمات المضادة للأكسدة مع تخفيض مستوى الشق الحر لأكسيد النيتروز إلى ذاك المستوى الموجود في المجموعة الضابطة.
كذلك فإن IP6 ساهم في زيادة العدد الكلي لخلايا الدم البيضاء و نسبة الخلايا الليمفاوية، بينما كان هناك نقص غير معنوي في نسبة كل من الخلايا المتعادلة و وحيدة النواة.
الخلاصة أن الـ IP6 له تأثير واضح و قوي على الحد من نشاط الشوارد و إفرازها و كذلك تحسين الجهاز المناعي في الجسم و تمكينه من التحكم في العدوى و القضاء على البكتريا الممرضة.
Abstract EN
Objective: The purpose of this study was to determine if inositol hexaphosphate (IP6) enhances the immune response and disease resistance against infections due to opportunistic pathogens Aeromonas hydrophila in male albino mice.
Matertial & Methods: Mice were classified into three groups: first group remained as a control group.
The second group was injected (i.p.) with A.
hydrophia once a week for four weeks at a dose of 0.2 ml of A.
hydrophia suspension (2 x 106 bacteria).
The third group was injected with A.
hydrophia suspension in the same way as the second one and synergistically IP6 was administrated three times a week for four weeks at a total dose of 40 mg/ kg mice.
Phagocytic activity, electrophoretic pattern of proteins and immunoglobulin levels were investigated.
Also, activities of the respiratory burst enzymes and nitric acid synthesis were estimated.
Results: Results of the second group revealed a marked increase in phagocytic activities, nitric oxide synthesis and a decrease in hepatocyte catalase, total peroxidase and superoxide dismutase activities.
On the other hand, in the third group, administration of IP6 enhanced the antioxidant enzyme activities.
At the same time, nitric oxide synthesis was shown to be in a similar rate to that of the control mice.
Furthermore, IP6 increased the total blood leukocyte counts and the proportion of lymphocytes, but it had a non-significant decrease of the proportion of neutrophils and monocytes.
Conclusion: The toxicity of free radicals produced by the bacteria was strongly reduced by IP6 administration.
