تم في هذا البحث فصل خمسة مركبات فلافونيدية-امنتوفلافون-رامنتين-كامفيرول-3-جلاكتوسيد-ليوتيولين-7-جلوكوسيد-كيرسيتين-3-جلوكوسيد من الخلاصة الكحولية و تم التعرف عليهم باستخدام طرق التحليل الطيفية المختلفة.
كما تمت دراسة الفاعلية البيولوجية للمركب الفلافونيدى امنتوفلافون كمضادات للخلايا السرطانية و قد اعطى هذا المركب فاعلية ايجابية على بعض اورام خلايا المخ بالمقارنة بعينة قياسية سيسبلاتين .
وتم ايضا فصل الزيت الطيار من اوراق نبات تاكسوديم والتعرف على مكوناتة بواسطة كروماتوجرفيا الغاز المتصل بطيف الكتلة ، ووجد ان التربينين 4-اول يمثل النسبة الاعلى في التواجد (37 و 38%) يليه مركب الفابينن و يمثل (51 و 14 %).
و بدراسة الفاعلية البيولوجية للزيت الطيار لهذا النبات كمضاد للفيروسات من نوع الهربس سمبلكس وجدانة اعطي فاعلية كبيره و تمثل حوالي ( 68 و 74 %).
Abstract EN
Objectives: To isolate flavonoids and volatile oil of Taxodium distichum leaves and evaluate the cytotoxic activity of the biflavonoid, amentoflavone against three types of tumor cell lines and evaluate the antiviral activity of the oil against Herpes simplex virus type 1 (HSV-1).
Methods: The dried powdered leaves of Taxodium distichum was extracted with 80% MeOH and then successfully extracted with pet.
ether (40-60 0C) and ethyl acetate.
Five flavonoids were isolated and identified by spectral analyses as, amentoflavone (1), rhamnetin (2), kaempferol-3-0-galactoside (3) luteolin-7-0-glucoside (4), quercetin-3-0-glucoside (5).
Cytotoxic activity of the isolated amentoflavone was performed against three types of tumor cell lines at a concentrations 1.00-10.00 ?g / ml using sulforhodamine B assay (SRB).The volatile oils of the fresh leaves was prepared by hydrodistillation method using Clevenger apparatus and the chemical composition of the oil was determined by GC/MS analysis .
The antiviral activity of the oil was tested against Herpes simplex virus type 1 (HSV-1).
Results: The tested compound exhibited a significant cytotoxic activity for U251, brain tumor cell lines (IC50 = 4.5 ?g / ml) while the IC50 was > 10 ?g/ml for both Hela and HEPG2 cell lines.
The major components of the volatile oil were terpinene-4-ol (38.37%) - pinene (14.51%), - terpinene (7.82%) and - terpineol (7.51%) and its antiviral activity against (HSV-1) was (74.68%) determined by plaque reduction assay method.
Conclusion: There is a potential cytotoxic activity of amentoflavone against brain tumor cell lines and significant antiviral activity of the volatile oils
