تهدف هذه الدراسة بحث إمكانية عزل الحمض النووي الريبوزي لفيروس الالتهاب الكبدي (C) باستخدام تقنية البلمرة المتسلسلة من عينات تم تجميعها و تجفيفها على نوع من ورق الترشيح المعامل (FTA) و كذلك مقارنة نتائج عزل الحمض الريبوزي للفيروس من عينات الدم الجافة على ورق الترشيح DBS)) مع عينات الدم المحفوظة بالتجميد.
تم تجميع عينات دم وريدي من الأشخاص المتطوعين الخاضعين لهذه الدراسة و هم عبارة عن 15 مريض بالفيروس الكبدي من ذوي التحليل الإيجابي في المصل (الأعمار من 25 – 62عاما) و كذلك خمسة متطوع (كحالات حاكمة) ليس لديهم إصابة بالفيروس.
وقد تم تنقيط 50 ميكروليتر دم (من نفس الأشخاص) على ورق ترشيح ((FTA Cards عن طريق وخز الإصبع ثم تجفيفها بهواء الغرفة لتصبح بذلك قادرة على التعامل بتقنية البلمرة المتسلسلة مباشرة.
أشارت النتائج إلى أن الحامض النووي الريبوزي للفيروس الكبدي C)) كان ثابتا في ظروف التجفيف و أمكن عزله من الهينة، و أشارت المقارنة بين نتائج التحليل بتقنية البلمرة المتسلسلة للعينات المحفوظة بالتجميد و تلك المأخوذة على ورق الترشيح (DBS) .
لنفس المرضى إلى التوافق التام في النتائج فكرة هذه الدراسة تقوم على تأسيس وسيلة جديدة في التحاليل المختصة بالتقنيات الحزيئية بمعنى آخر تهدف إلى إيجاد تحسن ملحوظ في طرق تجميع العينات فيما يخص نقلها من مكان إلى آخر، لا سيما من الأماكن البعيدة إلى المعامل المركزية ليتم تحليلها بتقنيات تحديد كمية الحامض النووي حتى و لو لم تتوافر ظروف التجميد.
Abstract EN
THE OBGECTIVE: The aim of this study was the investigation of recovering hepatitis C virus (HCV), RNA (Ribonucleic acid) using PCR technique (Polymerase chain reaction) from filter paper (FTA, Flinders Technology Associates), as well as, comparing results of using dried blood spotted samples on filter paper (DBS) with blood samples of the same patients in order to verify the efficiency of using (DBS).
MATERILS AND METHODS: Blood samples were collected from the subjects of this study (fifteen seropositive for anti-HCV patients; volunteers, with age range from 25 to 62 years and five volunteers control who were HCV antibody-negative).
50?l samples finger prick blood were blotted onto filter paper (FTA cards) from each subject allowed to air dry before processing for PCR .Another 5ml venous blood samples were collected from the same subjects (for the comparative study).
DBS were stored at room temperature before processing for PCR assay.
RESULTS: Results showed that HCV RNA was stable in a dried condition and could be recovered.
Comparison of PCR results of blood samples and DBS of the same patients showed complete matching.
CONCLUSION: The concept of this study was establishment of a new surveillance tool for molecular techniques i.e.
will constitute a significant improvement in the collection of samples in the field and transport, especially from remote areas of the world to centralized laboratories and analysis by nucleic acid amplification techniques even when freezing conditions are not available.
