Arabic Abstract

تعتبر هذه الدراسة الأول من نوعها في الحصول على بيانات بالاعتماد على الترددات القصيرة المنتشرة على Y كروموسوم (Y-STR’S), حيث تم تحديد التردد لـallele و التردد لنمط الجيني و التردد لأقل نمط جيني مشترك minimal haplotype في مجتمع شمال الأردن, لسبعة عشر موقع على Y كروموسوم (DYS456-DYS389 I - DYS390-DYS389 II-DYS458-DYS19 / 394-DY0S385 a / b- DYS393-DYS391-DSY439-DYS635-DYS392- H4-DYS437 - DYS438 -DYS448).

حيث تم عزل المادة الوراثية من 43 ذكر ليس بينهم أي درجة قرابة و تحليلها باستخدام ,multiplex polymerase chain reaction (PCR) و تم تحديد النمط allele باستخدام flouoresent detector of an automated DNA analyzer (310 Genetic analyzer).

من النتائج لوحظ أن تكرار المقاطع تفاوت من 0 إلى 31 لسبعة عشرة موقع, حيث ظهر 35 نمط جيني مختلف بتوزيع 0.991 و كان النمط الجيني المشترك بين العينات يتكون من ثلاث مواقع (DYS392-DYS437-DYS438) Y-STR’s, و تعد هذه النتائج التي تم الحصول عليه أداة جيدة لتميز مجتمع شمال الأردن و خطوة أساسية في تأسيس قاعدة بيانات للجينات الأردنية.

English Abstract

This study reports the first data on Y- chromosome specific short tandem repeats (STR's), allele frequencies, haplotype frequencies and minimal haplotype for seventeen Y-STR's locus (DYS456-DYS389 I - DYS390-DYS389 II-DYS458-DYS19 / 394– DY0S385 a / b-DYS393-DYS391-DSY439- DYS635-DYS392-H4-DYS437-DYS438-DYS448), were determined in population from Northern of Jordan.

DNA samples from 43 unrelated males were analyzed by using the multiplex polymerase chain reaction (PCR), the allele patterns were detected by the fluorescent detector of an automated DNA analyzer (GA310, Applied Bio system, USA), The observed number of alleles varied from 0 to 31 alleles per locus, from the data all seventeen loci were highly polymorphic.

A total of 35 different haplotypes were identified by the seventeen Y-STR loci.

The overall haplotype diversity was 0.991.

Wherever the minimal haplotype is composed of three Y-STR's (DYS392-DYS437-DYS438), which were observed in the Northern of Jordan samples.

In conclusion the data of allele frequency for the whole loci, considered a good tool to identify Jordanian and also the primary step to establish a primary DNA data base.
