English Abstract
Growth conditions which were expressed as biomass or OD6oo nm as well as the production of (3-galactosidase enzyme were investigated in different strains of Enterobacter aero genes.
Three Enterobacter strains were included in this studies and there are : pUC8 :16 ; A transform ant carrying Vitreoscilla hemoglobin gene (ygb) and two control strains.
The first (pUC9) is a transform ant carrying the plasmid only without vgb while the other control represents parental cells.
The optimum growth conditions were not totally identical to these for the enzyme production.
PH 7 and agitation rate of 150 rpm were common for both measurements.
However the optimum temperature for growth was approximately 37°C while that for (3-galactosidase synthesis was approximately 42°C.
The latter temperature gave almost five fold higher rate of enzyme production over the 37°C.
The bacterial growth was relatively stable within a range of 0.08-0.5 % NaCl supplement while the enzyme synthesis was severely repressed.
The effect of five different nitrogen sources (three organic and two inorganic) was also investigated either by using various concentrations of each nitrogen supplement or under fixed concentration of different nitrogen sources.
It was found that all used nitrogen sources either induce or have little effects on bacterial growth.
In contrast the p-galactosidase production was repressed in all Enterobacter strains through more severely in presence of inorganic nitrogen supplements.
The VHb protein in the transformed Enterobacter pUC8 : 16 strains had shown a protective action by minimizing the enzyme repression caused by salt or nitrogen supplements.
