English Abstract

-Three different flocks of Awassi sheep were tested for their genetic variation in south Jordan.

Ear tissue samples were collected from ewe, rams and lamb by Ear Puncher.

The total number of sheep from of the three flocks was 140 samples.

Those flocks were from Hu sinieh 1 (H1), Husinieh 2 (H2) and Al-Shoubak (SH) from Maan district.

The sample genomic DNA was extracted, quantified and amplified PCR for six Microsatellite markers (MS).

The six ovine and bovine MS, located on chromosomes 2 and 6 of sheep genome, were employed in genotyping experiments to produce PCR products then visualized using Silver Sequence™ DNA System.

The visualized bands were scored as allele size for each MS then genetically analyzed for allele frequency, expected and observed heterozygosity and polymorphism under HardyWeinberg equilibrium (HWE) by Cervus® software package.

On the other hand genetic distances between flocks were estimated using GDA® software package.

High genetic variation in the three flocks was found and revealed by measuring the heterozygosity measures, polymorphism and the number of alleles per locus.

The latter showed specific pattern of allele frequency profile in which some alleles found in one flock but not in another as well as in sometimes was of high frequency.

This result of allele profile can be considered for different genetic studies.

In particular, allele frequency profile was useful tool to explain the level of genetic differentiation between flocks.

Genetic distances were also undoubtedly revealed the expected degree of differentiation in the three flocks.

However, the three flocks comply well with HWE, in spite of they are small in effective population size.

Though mating was random with respect to loci in the study and both gene flow and selection pressures were not effective in changing the equilibrium state.

Finally This study of molecular and population genetic has provided the first knowledge and information of some genetic population parameters of sheep flocks in Al-Shoubak in South of Jordan.

To sum up, the outcomes of this study are expected for MS loci, which demonstrate high level of polymorphism so far.
