English Abstract

Purpose of the present study was to determine the effects of splenectomy and dacarbazine drug on some hematological and biochemical parameters in male albino rats.

The present study divided into two main experiments, the first experiment includes the study of the effects of splenectomy and the second experiment includes the study of the effects dacarbazine drug, the study was performed on seventy six male albino rats.

The rats divided in to two main experiments, the first experiment includes eight groups (control, sham, splenectomy, splenectomy dacarbazine, + Vit.

B12, + EPO, + Folic acid and combination group).

The second experiments also divided in to five groups (control, dacarbazine, dacarbazine + Vit.B12, + EPO and Folic acid).

The following are the most important results were obtained from the present studies.

1- The results of the first experiment showed that : A-In splenectomized rats; RBCs, Hb, MCHC, AST, ALT, ALP and total bilirubin were significantly decreased while, WBCs, Lymphocyte %, PLTs, Reticulocyte, serum iron, serum ferritin and serum MDA were significantly increased when compared with control.

B-Intraperitonealy injection of dacarbazine drug (4.5mg / Kg body weight) in splenectomized rats, significantly decreased RBCs, Hb, reticulocyte, PCV, and lymphocyte %, while, AST, ALT, total bilirubin, serum ferritin and serum MDA were significantly increased when compared with splenectomized rat group.

C-Intramuscular Injection of vit.B12 (500 mcg / kg body weight) in splenectomized rats, significantly increased reticulocyte, while, serum iron and MDA significantly decreased when compared with splenectomized rat group.

D-Subcutaneous injection of rHuEPO (100 IU / Kg body weight) in rats, significantly increased reticulocyte while, ALT, serum iron and MDA were significantly decreased.

E-Orally administration of folic acid (60 mg) per each kg diet in splenectomized rats increased reticulocyte while, serum iron and serum MDA significantly decreased when compared with splenectomized rat group.

F-In combination group (Splenectomy + Vit.

B12 + EPO + Folic acid) RBCs, reticulocyte and PLTs, were significantly increased while, serum iron, MCV, MCH, ALT and serum MDA were significantly decreased when compared with splenectomized rat group.2- The results of the second experiment showed that : A-Intraperitoneal injection of rats by dacarbazine (4.5 mg / Kg body weight) of rats showed significant decrease in RBCs, WBCs, Hb, Reticulocyte, PLTs, PCV and Lymphocyte, while, AST, ALT, ALP, total bilirubin, serum iron, serum ferritin and serum MDA, were significantly increased when compared with control.

B-In (Dacarbazine + Vit.

B12) treated group the reticulocyte, PCV and WBCs were significantly increased while ; serum iron and serum MDA were significantly decreased in comparison to dacarbazine treated group.

C-In (Dacarbazine + EPO) treated group the RBCs, Hb, reticulocyte, WBCs, and PLTs were significantly increased while, AST, ALT, ALP, total bilirubin, serum iron, serum ferritin and serum MDA were significantly decreased in comparison to dacarbazine treated group.

D-In (Dacarbazine + Folic acid) treated group the WBCs and PLTs were significantly increased while ; serum iron and serum MDA were significantly decreased in comparison to dacarbazine treated group.

E-Intraperitoneal injection of dacarbazine caused obvious inflammation and cell death in the histological sections of the liver.
