In this study a total of 26 microorganisms were isolated for lipase production.

Yeast strain possessing the highest lipase activity was identified as Sporobolomyces salmonicolor, and subjected to mutagenesis to increase the enzyme production.

Effect of pH, temperature and medium composition on lipase production was studied.

Sporobolomyces salmonicolor a novel yeast was isolated from oil spillage and the production conditions were optimized.

The lipase activity was increased by mutagenesis and with the optimum conditions at pH 6 and 28 °C for 96 h.

The mutant yeast strain OVS8 was selected as it showed stability and maximum lipase production, which is 3.2 times greater than the wild strain (38.5 ± 0.21U ml-1).

A novel lipase producing yeast was isolated from oil spillage and the lipase activity was enhanced.

Use of Sporobolomyces salmonicolor OVS8 for lipase production may be advantageous over Bacteria and Candida.
