تم تصنيع اللبن الرائب (yogurt) من حليب بقري معامل بتراكيز مختلفة من إنزيم الترانس كلوتامينيز الحر والمقيد و المعزول من بكتريا Streptoverticillium mobaraense.
أدت هذه المعاملة إلى ربط بروتين الكازين تقاطعيًا مع بروتينات الشرش، و بالتالي تحسين خواص اللبن الناتج.
لوحظت أعلى قيمة للرقم الهيدروجيني و أقل حموضة تسحيحية و خلال ثلاثة أسابيع من الخزن بدرجة حرارة º4م في العينة المعاملة بالإنزيم الحر بتركيز 75 وحدة/لتر.
أدت إضافة الإنزيم إلى الحليب في نفس وقت إضافة البادئ إلى زيادة معنوية في صلابة ولزوجة المنتج مع انخفاض في كمية الماء المفصول، و استمر هذا التأثير خلال الخزن.
كان تأثير إضافة 45 وحدة/ لتر مشابهًا لتأثير إضافة % 3 من الحليب الفرز المجفف.
كانت المعاملة بالإنزيم المقيد أقل تأثيرا" في صفات اللبن من المعاملة بالإنزيم الحر.
أظهر التقييم الحسي للعينات المعاملة بالإنزيم تفوقا" على العينات غير المعاملة في جميع الصفات الحسية و خلال جميع مدد الخزن ما عدا صفتي النكهة و الطعم الحامضي و خلال الأسبوع الأول فقط من التخزين.
أجريت عملية الترحيل الكهربائي بوجود العوامل الماسخة (SDS-PAGE) لبروتينات اللبن المعامل بالإنزيم المذكور، و بينت النتائج ظهور حزمة جديدة ذات وزن جزيئي عال ناتجة عن الربط التقاطعي لبروتينات الحليب بتأثير إنزيم الترانس كلوتامينيز، مع انخفاض في كثافة حزم بروتينات الكازين و بروتينات الشرش.
Abstract EN
Yogurt was manufactured from cow's milk which was treated with different concentrations of free and immobilized Streptoverticillium mobaraense transglutaminase.
The highest pH and lowest titratable acidity during three weeks of storage were detected in the sample treated with 75 U/ l of free transglutaminase before starter addition.
The concomitant MTGase treatment with starter addition resulted in significant increase in yogurt gel strength and viscosity and decreased water separation.
This effect was continued during storage at 4 The effect of enzymatic treatment with 45 U/ l almost resembled that of adding 3 % of C◦.
skimmed milk powder.
Immobilized MTGase treatment had less effects on yogurt properties.
Sensory evaluation of yogurt showed that treated samples were superior to untreated sample in all sensory aspects through storage periods.
To reveal the occurrence of cross-linking among protein molecules, SDS-PAG electrophoresis was performed for yogurt proteins.
New high molecular weight protein bands had occurred accompanied by decreasing in casein and whey proteins bands density.
