منذ بداياته قبل عقدين من الزمن و مع التحليل الكروموسومي لنقاط كسر الإرسال، و البحث في الآلية المرضية لسرطان الدم الليمفاوي الحاد في تقدم الآن على نطاق واسع في تسلسل الجينات المرشحة و التي قد تكون مرتبطة بالتسبب في سرطان الدم.
و قد استرعى الأنترلوكين -15 اهتمام العديد من متخصصي الأورام إلى خمسة من عديدي الشكل النووي المنفرد.
و التي ثبت علاقتها بسرطان الدم الليمفاوي الحاد في الأطفال.
و كان الهدف من هذه الدراسة بحث العلاقة بين الأشكال المتعددة لجين الأنترلوكين -15 و خطورته للبالغين المصابين بسرطان الدم الليمفاوي الحاد و عما إذا كانت ترتبط هذه الأشكال المتعددة لنمط ظاهري مناعي للمرض.
و شملت هذه الدراسة 60 شخص مقسمين إلى مجموعتين : 30 مريض الذين يعانون من سرطان الدم الليمفاوي الحاد للبالغين و 30 شخصا سليما من العمر و الجنس، كمجموعة ضابطة.
و لكل الأشخاص تم فحص النوع الجيني للrs10519613 و rs35964658.
و لكن كان هناك 1 و 2 أضعاف زيادة خطر سرطان الدم الليمفاوي الحد للبالغين في C-أليل الناقلين من rs10519613 تعدد الأشكال (أو : 2.1 95 % Cl : 0،45-9،84(.
بخصوص النمط الظاهري المناعي للمرض، لم يكن هناك فرق إحصائي بين نوع خلية-بي و نوع خليه-تي بشأن توزيع الموروثات من الأشكال الاثنين، و لكن هناك 1.2 أضعاف لخطر متزايد للخلية-بي في النوع حاملي G-أليل لل rs35964658 تعدد الأشكال (أو : 1.2 95 % : 0،07-19،63(.
و خلص إلى أنه لا يوجد أي ارتباط بين الأشكال المتعددة لل للrs10519613 و rs35964658 على حد سواء، و سرطان الدم الليمفاوي الحاد للبالغين و لا النمط الظاهري للمناعة من المرض.
Abstract EN
Autophagy is originally described as the main catabolic pathway responsible for maintaining intracellular nutritional homeostasis that involves the formation of a unique vacuole, the autophagosome, and the interaction with the lysosome.
Recent evidence indicated the essential role of the host autophagy, in enhancing influenza A virus (IAV) replication in human lung epithelial cells.
Here, findings demonstrate that IAV infection increased levels of the autophagosome marker ``microtubule-associated protein light chain 3-II`` (LC3-II), at early stage of infection.
Further, knockout of Atg5, the crucial components of autophagosome formation, and siRN Amediated depletion of autophagy related genes Atg9, Atg12, Atg16 provide further evidence on the essential and supportive role of autophagy in IAV replication.
Interestingly, in autophagy-deficient cells (Atg5- / - MEFs), and A549 cells lacking autophagy levels of host protective type I interferon's, in particular IFN-, were increased during infection.
Taken together, these data further confirm the beneficial role of autophagy for IAV infection may be via regulation of IFN- production and highlights autophagosome formation as a potential novel antiviral target.
