الهدف من هذه الدراسة هو تقييم التعبير عن بعض الجينات مسارنوتش، بالإضافة إلى عامل -1 ألفا المحرض بنقص الأكسجة خلال تسرطن الخلايا الكبدية في الفئران.
و هذا يلقي الضوء على دور التعبير عنها في سرطان الكبد.
و علاوة على ذلك، فإن الدراسة تهدف إلى تحديد أي علاقة متبادلة بين إشارات نوتش و نقس الأكسجين في تطور سرطان الكبد في الفئران.
و قد تم استحداث التسرطن الكبدي كيميائيا في فئران ذكور.
و تم فحص عينات الأنسجة و تصنيفها وفقا لذلك إلى مجموعة مبكرة سابقة للورم و مجموعة متقدمة سابقة للورم، و مجموعة ضابطة.
و تم تقييم التعبير عن جينات نوتش 1، دلل 4-، عامل -1ألفا المستحث بنقص الأكسجة في عينات الأنسجة عن طريق استخراج الحمض النووي الريبي الكلي و إجراء تفاعل Reverse Transcriptase-PCR.
و قد لوحظ زيادة كبيرة في التعبير عن الجينات المستهدفة في المجموعات المريضة عنها في المجموعة الطبيعية.
كما كان هناك ارتباط كبير بين التعبير عن نوتش 1، دلل 4- عامل -1ألفا المستحث بنقص الأكسجة.
و من ثم يمكن استنتاج أن زيادة التعبير عن كل من نوتش 1، دلل4-، و عامل -1ألفا المستحث بنقص الأكسجة ترتبط بشكل وثيق مع التحول السرطاني لخلايا الكبد، كما أنه من الممكن وجود علاقة بين نوتش و نقص الأكسجة قد تسهم في تسرطن الخلايا الكبدية.
Abstract EN
Hepatocellular carcinoma (HCC) is a common worldwide malignancy.
Notch signaling pathway contributes to the genesis of diverse cancers, however, its role in HCC is unclear.
Hypoxia is a common feature of HCC.
Signal integration between Notch and hypoxia may be involved in HCC.
The aim of this study was to evaluate the expression of some Notch pathway genes, in addition to hypoxia inducible factor-1 alpha (HIF-1) during rat hepatocellular carcinogenesis.
The study aimed also, to identify any crosstalk between Notch signaling and hypoxia throughout hepatocellular oncogenesis in rats.
Hepatocellular carcinogenesis was chemically induced in rats.
Tissue specimens were examined histopathologically and classified into early preneoplastic group, advanced preneoplastic group, and control group.
The expression of Notch-1, Dll-4, and HIF-1 was evaluated in tissue samples by total RNA extraction and RTPCR The expression of target genes were significantly increased in the diseased groups than in control group.
There was a significant correlation between the expression of Notch1, Dll-4, and HIF-1 .
These results indicate that overexpression of Notch-1, Dll-4, and HIF- 1 may be associated with the malignant transformation of hepatocytes with a possible Notch – hypoxia cross talk during hepatocellular oncogenecity.
