يهدف هذا البحث إلى دراسة التأثير الوقائي لعديدي التسكر المستخلص من الوسط الغذائي لميسيليوم فطر عيش الغراب المحاري ضد عقار الديكسوروبسين المسئول عن حدوث قصور في وظائف القلب في بالجرذان.
و قد أظهرت النتائج أن لعديدي التسكر تأثيرا معنويا عند حقنه للجرذان قبل عقار الديكسوربسين, حيث عادت مستويات كلا من أنزيمات (CK, ALT & LDH) في مصل دم الجرذان إلى مستوياتها الطبيعية.
و قد أوضحت أيضا النتائج تحسنا معنويا في مستوى تأكسد الدهون (المالونالدهيد الثنائي, MDA) و مضادات الأكسدة الجلوتاثيون و السوبر أوكسيد ديسميوتيز و الكاتليز (GSH, SOD &CAT) في أنسجة القلب في الجرذان.
أيضا مستوى كلا من أكسيد النيتريك (NO) و معامل التنكرز - ألفا (TNF-α) انخفض في أنسجة القلب, بينما الانترفيرون - جاما (IFN-) لم يظهر تغييرا ملحوظا مقارنة بالمجموعة الضابطة normal (control).
و لقد أظهر فحص خلايا عضلة البطين الأيسر للقلب تحت الميكروسكوب الضوئي عد وجود تغيير في التركيب البنائي للخلايا, ما عدا نقصان طفيف في سمك الألياف العضلية الدقيقة في بعض الأجزاء.
من نتائج هذا البحث يمكن استنتاج أن المعالجة بعديدي التسكر له تأثير وقائي لعضلة القلب من الآثار الجانبية الناتجة من استخدام عقار الديكسوربسين في الجرذان.
Abstract EN
The present study was undertaken to investigate the possible abatement of doxorubicin-induced cardiomyopathy in the rat by polysaccharopeptides extracted from the culture broth of Pleurotus ostreatus mycelium.
It was found that polysaccharopeptides given as protection before doxorubicin intoxication normalized the levels of serum total creatine kinase, aspartate aminotransferase and lactate dehydrogenase activities, in addition to malondialdehyde, reduced glutathione, as well as superoxide dismutase and catalase activities in the heart tissue.
Nitric oxide and tumor necrosis factor-α concentration were persistently reduced in the heart tissue, while interferon-γ concentration was insignificantly changed, compared to controls.
Light microscopic examination of Haematoxyline and eosin-stained sections of the left ventricles showed normal structure of the cardiac myocytes, except for a slight but significant decrease in their thickness, compared to controls.
In conclusion, pretreatment with mycelia polysaccharopeptides provided a potential protective effect against doxorubicin-induced oxidative stress in rats.
