Determination of perindopril erbumine in tablets by spectrophotometry

Perindopril erbumine (PDE), used in the treatment of patients with heart failure, belongs to a class of drugs known as angiotensin-converting-enzyme inhibitors.

Four (M1-M4) sensitive, accurate, precise and cost-effective spectrophotometric methods for the determination of PDE in bulk and in tablets dosage forms have been developed and validated.

Method M1 and M2 are based on the formation of chloroform soluble ion-pair complexes of PDE with safranin O and bromothymol blue in basic and acidic medium, respectively.

Method M3 is based on the oxidation of the PDE with alkaline potassium permanganate.

Method M4 involves the ternary complex formation of the PDE with molybdenum (V) thiocyanate.

The colored species showed absorbance maxima at 520, 410, 605 and 750 nm for methods M1, M2, M3 and M4, respectively.

The optimum conditions for all the four methods are optimized.

Beer’s law is obeyed over the ranges 5–25 μg ml-1 (M1) and 2-20 μg ml-1 (M2, M3 and M4).

The apparent molar absorptivity, Sandell’s sensitivity, limit of detection and limit quantification values are reported.

All the methods were validated in accordance with ICH guidelines.

The developed methods were employed with high degree of precision and accuracy for the estimation of drug content in commercial tablets of PDE.
