تم استخدام اختبار تفاعل البلمرة المتسلسل للتعرف على ميكروب القولون المعدي المفرزة للسموم المعوية سواء كانت معزولات حقلية أو مرجعية مستخدمة في تحضير التحصين المحلي و ذلك نظرا لحساسية و دقة التحديد المعروفة لهذا الاختبار و تم استخدام البادئ الجيني eaeA للكشف عن أنتيجين الأنتيمين Intimin الموجود في هذه النوعية من البكتيريا.
أيضا تم عمل الاختبارات الكيميائية و السيرولوجية لعدد أثنى عشر عترة من مجموعة مائة حالة مصابة.
تم تعريف المعزولات و العترات المستخدمة في هذه الدراسة باستخدام الاختبارات السابقة على أنهما E.
coli O157:H7، E.
coli O127:H6.
كما أوضح اختبار PCR أعلى نسبة توافق بين العترات المعزولة و العترة المستخدمة في إنتاج اللقاح.
Abstract EN
Enterotoxigenic E.
coli (ETEC) is the cause of calf morbidity and mortality in addition to zoonotic importance.
Vaccination against these causes is strictly required.
STEC isolates were isolated from faecal calf samples, biotyped according to biochemical tests and serotyped.
STEC genes amplified and detected through PCR.
Local vaccinal strain (K99) was included in PCR studies.
Serotypes O157:H7 and O127:H6 were commonly isolated from a field isolates.
Intimin gene was amplified and detected in all isolates, field isolates and vaccinal strain showed the same size band.
