English Abstract
Our study was designed for investigation the effects of newly prepared triazoloquinolone on male fertility model at University of Jordan.
Triazoloquinolone 9-Cyclopropyl-4-fluoro-3-(4-methoxy-phenyl)-6-oxo-6, 9-dihydro-3H-[l, 2, 3] triazolo [4, 5-hquinoline-7-carboxylic acid was chosen for the present study due to its activity angst human breast cancer cell lines and colorectal.
The Lethal Dose (LD50) of the triazoloquinolone compound was found to be 2000 (jig / kg) body weight.
Based on the LD50, three doses were used : 100,150 and 200 go / kg rat to study the effect of the triazoloquinolone on male rat fertility for sixty consecutive days.
The obtained results revealed that the triazoloquinolone compound treatment caused a significant (p < 0.05) reduction in sperm count from 33.61 million / ml for control epididymis suspension to 2 and 5, 8 million / ml epididymis suspension in rats which received 100, 150 and 200 / kg, respectively.
This was accompanied with a marked reduction in testes and epidedemes weight in treated groups in comparison with control.
A marked decrease was noted in cauda epididymal sperm motility with percentage of 83 %, 44 % and 31.46 % for 100, 50 ل and 200 / ig / kg, respectively, compared to the control 88%.
A significant reduction in testosterone concentration with values of3.52ng/ml for control group and 1.9, 1.6 and 1.5 ng / ml serum for 100,150 and 200 ير /kg, restively.
A significant reduction in Follicular Stimulating Hormone concentration from 0.098 iu / ml for control group to 0.036, 0.021 and 0.015 iu1 m serum for 100, 150 and 200 fig / kg, respectively.
The effect of oazolo quinolone compound was dose (pendent).
Histological examination of testes showed swelling and infiltration of iammatory cells into seminiferous tubules.
Immunohistological examination showed significant expression of both HSP70 and INOS in treated groups compared with control group (p < 0.05).
Conclusion : the toxicity of tested triazoloquinolone was investigated and shown to adversely affect the male reproductive system through induction of inflammatory changes in testes and increased expression of INOS.
Although triazoloquinolone compound has good activities against cancer cells, it is a compound with potential infertility effects.
