الغرض من هذا البحث هو لتقییم مدى إمكانیة استعمال تفاعل البولیمریز المتسلسل Polymrase chain (PCR) reaction في تشخیص بكتریا Pseudomonas aeruginosa المعزولة من جروح الحروق الملوثة، إذ تم التشخیص بمضاعفة 396-bp في تتابع القواعد النیتروجینیة للمورثة التركیبیة للذیفان الخارجي أ (exotoxin A) باستعمال بادئات خاصة بتلك المورثة .
أظهرت النتائج إن بكتیریا P.
aeruginosa الحاویة على مورثة الذیفان الخارجي (أ) أعطت نتیجة موجبة في الفحص أما بكتیریا Pseudomonas التابعة لنوع آخر غیر aeruginosa الغیر حاویة على مورثة الذیفان الخارجي (أ) أعطت نتیجة سالبة لهذا الفحص و أیضا تم اختبار خصوصیة و حساسیة هذا الاختبار و كانت النتائج 100 و %96 على التوالي و هذه النتائج تعزز إمكانیة استعمال هذا الاختبار في الدراسة التشخیصیة و الوبائیة لهذه البكتیریا و بینت النتائج إنه یمكن الكشف عن pg 0.2 من الحمض النووي الدیوكسي رایبوزي (DNA) لكل 3 مایكرولیتر من خلیط التفاعل إذ ظهرت النتائج الموجبة بشكل حزمة (DNA) بحجم 396-bp في هلام الاكاروز المرحل كهربائیا و المصبوغ بصبغة برومید الاثیدیوم.
و أثبتت الدراسة إن هذه الطریقة أسرع و أدق من الطرق التقلیدیة المتبعة لتشخیص هذه البكتیریا إذ یمكن الكشف من خلالها عن كمیات ضئیلة من البكتیریا في العینات المرضیة دون الحاجة إلى أوساط مزرعیة انتقائية أو اختبارات كیموحیویة إضافیة.
Abstract EN
The purpose of this research was to evaluate the use of PCR for the identification of burn wounds isolates of Pseudomonas aeruginosa by amplifying a 396-bp region of the exotoxin A (ETA) structural gene sequence.
Specific primers amplified ETA-positive P.
aeruginosa DNA, whereas other species of Pseudomonas and bacteria other than Pseudomonas which were isolated from burn wounds infections did not yield any 396-bp fragment.
The specificity and sensitivity of the assay were 100 and 96%, respectively, which confirms the assay's reliability for diagnostic and epidemiological studies.
The assay can detect 0.2 pg of P.
aeruginosa DNA per reaction mixture (3ul) by ethidium bromide staining of an agarose gel.
This method is rapid and less cumbersome than other diagnostic methods for the identification of P.
aeruginosa strains.
The method described can be used to detect a low level of P.
aeruginosa from clinical samples without the use of selective media or additional biochemical tests.
