استخدمت في هذه الدراسة ثلاث عزت من بكتريا EScherichia coli و التي جمعت من مصادر مختلفة تضمنت عينات من أنسان و فضلات الحصنة و التربة، أعطيت لها الرموز BO1، FH2، SL3 .
تم تضخيم الجين SSrA العائد لكل عزلة باستخدام تقنية PCR- co1onY و حصلنا على حزمة دنا واحدة لكـل عزلـة على هلام اكاروز.
دمج الجين SSr A المعزول من كل سلالة مع بلازمد pTrc 99A و كلون، و بعد هـضم المدمجات بأنزيمات القاطعة Eco R1 و Hind لوحظ وجود حزمتين من الدنا أحدهما مثلـت بلازما pTrc 99A و كانت بحجم 6 زوج قاعدي و مثلت الحزمة الثانية جين SSrA و كانت بحجم 400 زوج قاعدي.
فضلا عن ذلك تضمنت الدراسة قراءة للتسلسلات النيوكليوتيدية لجين SSrA للعزت الثلاثة و لوحظ وجود عدة طفرات شملت منطقة التشفير و المنطقة المحيطة بأعلى و أسفل مجرى الجين، و مـن خـلال هـذه الدراسة يمكن استنتاج أن جين SSrA غير محافظ.
Abstract EN
Three isolates of Escherichia coli, collected from different sources such as human specimens, feces of horses, and soil, designated as BO1, FH2, and SL3, were used in this study.
SsrA gene from each isolate was amplified using colony–PCR technique and one DNA band from each isolate on agarose gel was noticed.
Thereafter, SsrA gene was successfully combined with plasmid pTrc 99A and cloned.
Subsequently, the recombinants of pTrc 99A were digested with restriction enzymes, EcoR1 and HindIII.
The results revealed two bands ; one band represented plasmid pTrc 99A with 4000bp while the other represented SsrA gene with 600bp.
Furthermore, Sequence of SsrA gene of three isolates was done.
It was observed different mutations in SsrA gene involved the coding region as well as up–and down–stream of the flank regions of that gene.
From this study, it can be concluded that SsrA gene was not conserved.
