Abstract AR
الهدف من هذه الدراسة هو اختبار تأثير اكسندين -4 ( نظير للجلوكاجون شبه البيبتايد -1) علـى الـدورة الدمويـة فـي الفئـران البيضـاء المخـدرة باليوريثـان0.6 مـل /100 جـم.
و قـد أجريـت هـذه الدراسـة علـى 6 مجموعـات مـن ذكـور الفئـران البيضاء تتكون كل مجموعة من 6 فئران.
يتراوح وزنها بين 300-350 جرام.
المجموعة الأولـى و تشـمل الفئـران التـي تـم حقنها بالاكسندين -4 فقط في الوريد، المجموعة الثانية و قد تم حقنها بعقار البروبرانولول في الوريد و بعـدها ب 15 دقيقـة حقـن الاكسـندين -4 بـنفس الجرعـة، المجموعـة الثالثـة و قـد تـم حقنهـا بـالبروبرانولول بالإضـافة إلـى عقـار الفنتـولامين ثـم تبعها حقن الاكسندين -4 بعـد 15 دقيقـة فـي الوريـد بـنفس الجرعـة، المجموعـة الرابعـة تـم إجـراء الحرمـان العصـبي لنخـاع الغــدة الكظريــة و بعــد 15 دقيقــة تــم حقــن الاكســندين -4 فــي الوريــد بــنفس الجرعــة، المجموعــة الخامســة تــم حقــن هــذه المجموعة بالأتروبين في الوريد و بعد 15 دقيقة تم حقن الاكسندين -4 في الوريد و بنفس الجرعـة، المجموعـة السادسـة تـم حقن هذه المجموعة بعقار النيتروأرجينين ميثيل أستر و بعد 15 دقيقة تم حقن الاكسندين -4. و قد أجريت القياسات الآتية على جميع المجموعات سرعة ضربات القلب، متوسط ضغط الدم الشرياني، معـدل سـريان الدم في أوعية الأطراف الخلفية، المساريقا و الكلى.
و المقاومة في أوعية الأطراف الخلفية، المسـاريقا و الكلـى حيـث أظهـرت النتـائج زيـادة ذات دلالـة إحصـائية فـى سـرعة ضـربات القلـب و متوسـط ضـغط الـدم و اتسـاع الأوعيـة الدمويـة فـي الأطـراف الخلفية بينما نقصت سرعة سريان الدم فى المساريقا نقصا ذو دلالة إحصائية و هذا التأثير أبطـل بعـد حقـن البروبرانولـول و الفنتـــولاين و أيضـــا بعـــد الحرمـــان العصـــبيي لنخـــاع الغـــدة الكظريـــة بدلالـــة إحصـــائية إمـــا بعـــد حقـــن البروبرانولـــول أو النيتروأرجينين ميثيل أستر فإن سرعة سريان الدم في المساريقا نقص نقصا ذو دلالة إحصائية. و من هنا يتضح أن الاكسندين -4 له تأثير على القلب و الأوعية الدمويـة و فسـرت هـذه النتـائج علـى أسـاس التـأثير على تنشيط الجهاز السمبثاوي و نخاع الغدة الكظرية و كذلك من خلال مستقبلات جلوكاجون شبة البيبتايد-1.
Abstract EN
Aim : The aim of the present study is to investigate the hemodynamic effect of exendin-4 (Ex-4) on rats and its underlying mechanism. Material and Methods : six groups of male albino rats (6 each), group I: injected I.V with Ex-4 in a bolus dose of 250 ng / kg, group II: injected I.V with propranolol in a dose of 1 mg/kg 15 min before Ex-4, group III : injected with Propranolol in the same dose and phentolamine in a dose of 1 mg/kg.
15 min before IV injection of Ex-4, group IV: adrenomedullary denervation was done before IV injection of Ex-4, group V : atropine in a dose of I mg / kg I.
V.
15 min before Ex-4, group VI : L-NAME IV injected in a dose of 3 ng / kg 15 min before Ex-4 injection.
Heart rate, mean arterial pressure, hindlimb, mesenteric and renal flow and resistance were recorded. Results : IV injection of Ex-4 shows significant increase in heart rate, mean arterial pressure and hindlimb flow while the mesenteric flow significantly decreased and this action is abolished significantly after injection of propranolol and phentolamine also after adrenomedullary denervation while the increase in hind limb flow insignificantly changed after propranolol and L-NAME and the pressor action over mesenteric flow increased significantly. Conclusion : It can be concluded that Ex-4 has cardiovascular effects some of which include sympathoadrenal activation and others may be through GLP-1 receptor dependent action.
