تناولت الدراسة الحالية التعرف على وجود و فعالية أنزيم الثايميدين فوسفوريليز Tpase (EC.2.4.2.4) في الجزء الرائق من متجانس الرؤيسات الأولية للمشوكة الجيبية.
كما تمت دراسة بعض الخواص الحركية لهذا الإنزيم.
و وجد أن الديوكسي يوريدين يمكن أن يكون مادة أساس بديلة عن الثايميدين (المادة الأساس) أثناء التفاعل المحفز بأنزيم Tpase.
كما أظهرت النتائج أن لهذا الأنزيم حساسية عالية للتثبيط بالقاعدة النتروجينية الثايمين (ناتج التفاعل) و السايتدين و العقارين 5- فلورويوراسيل و البندازول بنسبة تثبيط 36 %، 68 %، 52 % و 3 % على التوالي.
إن وجود فعالية إنزيم الثايميدين فوسفوريليز يدل على وجود مسار الاسترداد (الإنقاذ) في تكوين نيوكليوتيدات الثايمين الضرورية لبناء الحامض النووي الرايبوزي المنقوص الأوكسجين DNA في الرؤيسات الأولية للمشوكة الجيبية، بالإضافة إلى المسار الحقيقي لتكوين هذه النيوكليوتيدات.
Abstract EN
The present investigation indicates the presence of thymidine phosphorylase (TPase) (EC.2.4.2.4) and its activity in supernatant fraction of Echinococcus granulosus protoscoleces extract, furthermore some of its kinetics properties were investigated.
It was found that deoxyuridine might be an alternative substrate for thymidine during catalyzed reaction by enzyme TPase.
The result also indicated that TPase activity was extremely sensitive to inhibite by thymine (product reaction), cytidine, 5-flourouracil and albendazole with inhibitory percentages of : 36 %, 68 %, 52 % and 3 % respectively.
The presence of TPase activity indicates the presence of salvage pathway for thymine nucleotide which was essential to deoxyribonucleic acid (DNA) synthesis in this organism, in addition to de novo pathway.
