الهدف : تهدف الدراسة الحالية إلى تحديد فعالية استخدام تقنية سلسلة تفاعل الإنزيم المتبلمر PCR في الكشف و التفريق بين أنواع طفيليات اللشمانيا الجلدية في العينات السريرية المأخوذة من المصابين بطفيلي اللشمانيا الجلدية.
المنهجية : تم فحص العينات المأخوذة من (115) مريض مصابين بالآفة الجلدية باستخدام المسحة الزجاجية و الزرع المختبري على الوسط الزرعي (NNN) و التكثير على الوسط الزرعي (RPMI 1640) و استخدام الوسائل الإحصائية من نسبة مئوية و تكرار.
النتائج : تم تضخيم الـ DNA في الطور أمامي السوط من العينات المأخوذة من الأوساط الزرعية باستخدام بادئات خاصة للجين المشفر الـ DNA للبائنة الحركية المسمى (Kinetoplast kDNA) لطفيلي اللشمانيا.
أظهرت الدراسة بأن هناك نوعين لطفيلي اللشمانيا الجلدية احدهما (L.
major) و وزنها الجزيئي (620) زوجا قاعديا و الأخرى (L.
tropica) و وزنها الجزيئي (800) زوجا قاعديا في العينات المجمعة من المصابين بهذا الطفيلي.
الاستنتاج : نستنتج من الدراسة الحالية أن استخدام تقنية الـ PCR ذا حساسية و خصوصية عالية عند التفريق بين أنواع اللشمانيا الجلدية عند مقارنتها بالطرق التقليدية و قد بينت النتائج المستحصلة من هذه التقنية أن هنالك نوعين من طفيلي اللشمانيا الجلدية للمصابين تم التحري عنها في محافظة النجف الأشرف.
التوصيات : استخدام تقنية الـ PCR في التشخيص و التفريق بين أنواع اللشمانيا الجلدية.
Abstract EN
Objective : The present study aimed to determine the effectiveness of a polymerase chain reaction (PCR) technique for identification and differentiation of the cutaneous leishmaniasis parasite in clinical sample collected from lesion exudates patients.
Methodology : the (115) sample were examined by smear slid preparation culture on RPMI 1640 and NNN then DNA isolation.
The DNA of the promastigote were amplified by PCR including primers selected on repetitive KDNA for identification of leishmania species.
The data was analyzed by using frequency and percentage.
Results : the PCR result showed that two species of leishmania (L.
major with 620 bp and L.
tropica with 800 bp) exist.
Conclusion : it is concluded from the present study that the PCR technique has high specificity and sensitivity during the differentiation between Cutaneous Leishmanial species in comparing with conventional methods.
Recommendation : the PCR technique can be use it to diagnosis and differentiation between different type of Cutaneou Leishmaniasis.
