أثبتت الدراسة وجود بكتيريا Bacillus thuringiensis في جميع النظم البيئية المختلفة، التي تم فحصها في سورية، حيث توافرت و بغزارة في الترب.
صنفت 219 (97%) عزلة لبكتيريا B.
thuringiensis المنتجة للبلورات البروتينية إلى 8 أنواع بيوكيميائية.
و كانت B.
thuringiensis subsp.
kurstaki الأكثر شيوعا.
و ظهرت 192 (88%) عزلة من أصل 219 عزلة سميتها ليرقات العمر الثالث لعثة طحين البحر المتوسط Ephestia kuehniella Zeller (Pyralidae: Lepidoptera).
و تبين أن العزلات التي عزلت من المناطق الزراعية و التي تحوي بلورات على شكل معين و مكعب هي الأكثر سمية.
أثبت وجود علاقة ما بين شكل االبلورات و فاعليتها ضد الحشرات.
فتبين أن العزلات التي تحتوي على بلورات ذات شكل معين أكثر سمية مقارنة مع العزلات التي تحتوي على بلورات ذات شكل كروي.
و استخدم تفاعل البلمرة المتسلسل (PCR) للتنبؤ عن مدى فاعلية العزلات المعزولة من التربة ضد الحشرات و مدى المطابقة في الفاعلية للعزلات السامة مع طريقة التقييم الحيوي.
حيث وجد أن العزلة الأكثر سمية لعثة طحين البحر المتوسط تحتوي على جينين مختلفين هما : cryI, cryIV التي تختص بفاعليتها السامة ضد الحشرات التابعة لرتبة حرشفية ورتبة ثنائية الأجنحة.
Abstract EN
Bacillus thuringiensis was found in all tested different ecosystems in Syria.
It was highly abundant in soils.
A total of 219 isolates (97 %) of B.
thuringiensis producing parasporal crystals was classified into eight biochemical types.
Bacillus thuringiensis subsp.
kurstaki was the most common.
Out of 219 strains, 192 strains (88 %) were toxic to the third instar larvae of Ephestia kuehniella Zeller (Pyralidae : Lepidoptera).
The highest toxic strains were isolated from cultivated areas and characterized by producing both bipyramidal and cuboidal parasporal crystals.
Relationship between crystal shape and toxic activity was observed.
The frequency of toxicity was greater in strains that produced bipyramidal parasporal crystals than strains that produced spherical crystals.
Polymerase chain reaction (PCR) analysis of bacterial isolates gave characteristic products that allowed rapid prediction of the insecticidal activity.
Insecticidal activity predicted by the PCR screen was found to correspond with the insecticidal activity of insect bioassay.
The most toxic strain harbored different specific cry genes including cryI and cryIV genes which are insecticidal for Lepidopteran and Dipteran insects.
