The ability of spinach beet, Beta vulgaris ssp, cicla (L.) to callus and to regenerate in vitro under normal and NaCl salinity stress conditions was investigated.

Seed halves as well as leaf and stem pieces were cultured on MS (Murashig and Skoog 1962) media supplemented with different levels and combinations of the plant hormones BA , IAA and NAA with and without NaCl.

Terminal bud explants were cultured on MS media supplemented with BA, in the absence and presence of NaCl.

Callus was obtained from leaf explants in the presence of 0.2 mg/L IAA plus 2.0 mg /L BA and 172 mM NaCl.

From stem explants, callus was obtained in MS media containing different levels and combinations of the plant hormones BA and IAA, in the presence and absence of 172 mM NaCl.

Shoots were produced from terminal bud explants after 2-3 weeks of culturing .

There was no significant difference in the production of shoots between the control and those treated with 35 and 86 mM NaCl in the presence of 0.25mg/LBA.

At 172mM NaCl however, shoots production increased significantly from 50% in the control to 71%.

Spinach beet plant has the ability for callus formation and in vitro regeneration under normal and NaCl salinity stress conditions.
