أجريت هذه الدراسة على النشاط المضاد للفطريات لعدد 65 عزلة من الأكتينوميسيتات ساكنة النبات و المعزولة من 12 نوع من النباتات الطبية البرية في مصر.
و قد تم تحديد نشاطها معمليا بإستخدام تقنية التضاد الحيوي بين المزارع المزدوجة ضد أربعة من الفطريات الممرضة للنباتات : فيوزاريوم سولانى Fusarium solani, و فايتوفثيرا إنفيستانس Phytophthora infestans, و ماكروفومينا فاسيولينا Macrophomina phaseolina, و بوترايتس سينيريا Botrytis cinerea.
و هي العوامل المسببة لذبول الطماطم, و اللفحة المتأخرة في البطاطس, و التعفن الفحمي للفاصوليا, و تعفن ثمار الباذنجان, على التوالي.
و قد أظهرت النتائج أن عزلتي ستربتوميسيس نوع 1 (Streptomyces sp.1) و كيتاساتوسبوريا نوع 5 (Kitasatosporia sp.
5) هما من أقوى العزلات في إنتاج مركبات مثبطة للفطريات.
و قد تم اختبار قدرة كلا من العزلتين على إنتاج مركبات مضادة للفطريات و مركبات متطايرة و إنزيمات (الكيتيناز و السليلوزات و كاربوكسيميثيل سيليولاز).
و قد أظهرت العزلتين نشاطا مختبريا ذي دلالة معنوية إحصائية (P < 0.05) ضد مسببات الأمراض الفطرية الأربعة سالفة الذكر, بالإضافة إلى فطر آخر هو فيوزيريام أوكسيسبوريام Fusarium oxysporum.
و قد وجد أن عزلتا الأكتينوميسيتات تستخدمان آليات مختلفة لمكافحة مسببات الأمراض, حيث كان التنافس على الغذاء في الوسط الغذائي الصلب أقل فعالية في تثبيط النمو الفطري, مقارنة مع تلك المسجلة عند استخدام نواتج الأيض المنتشرة في الأجار.
و قد تم تسجيل أكبر التأثيرات المضادة من خلال نواتج الأيض الأولية و نواتج الأيض النافذة من السيلوفان و المواد المتطايرة على التوالي, إلا أن كلا العزلتين لم تظهرا القدرة على إنتاج إنزيمات الكيتيناز أو السليلوزات.
و بناء على النتائج تم مناقشة إمكانية استخدام السلالتين معا كعوامل مكافحة بيولوجية للسيطرة على الفطريات الممرضة للنباتات.
Abstract EN
The present study is focused on the antifungal activities of 65 endophytic actinomycetes that were recovered from 12 wild medicinal plants in Egypt.
In vitro antifungal activity was determined using the dual culture bioassay against four fungal phytopathogens, Fusarium solani, Phytophthora infestans Macrophomina phaseolina and Botrytis cinerea, which are the causative agents of tomato's wilt, potato's late blight, soybean's charcoal rot and eggplant fruit rot, respectively.
Out of the 65 isolates, Streptomyces sp.
1 and Kitasatosporia sp.
5 were prolific producers of fungal inhibitory compounds.
Both isolates were investigated for the production of antifungal diffusible compounds, volatile compounds and enzymes (chitinase, cellulase and CMCase).
They showed in vitro activity against the four previously mentioned fungal pathogens as well as another pathogen (Fusarium oxysporum), with a probability of p < 0.05.
The actinomycete isolates employed various mechanisms for inhibiting the pathogens.
The competitive growth between pathogens and antagonists on solid media revealed lesser suppression, comparing to that recorded for the diffusates on agar.
Additionally, greater antagonistic effects were recorded for crude metabolites, membrane diffusible metabolite(s), and volatiles, respectively.
However, both actinomycete isolates did not show abilities to produce chitinases or cellulases.
The potential of using Streptomyces sp.
1 and Kitasatosporia sp.
5 as phytopathogen biocontrol agents was discussed.
