تم استحثاث بكتريا Klebsiella pneumonia (K64) المعزولة سريريا من عينة إدرار بوساطة مادة مطفرة (Mitomycim C) لإنتاج الكليبوسين.
تمت تنقية الكليبوسين باعتماد كروماتوغرافيا التبادل الأيوني.
تم الحصول على الكليبوسين بكروماتوغرافيا الترشيح الهلامي ؛ و رحل البروتين المنقى بال SDS-PAGE.
اختبرت الفعالية ضد المايكروبية تجاه أحياء مجهرية مختلفة شملت :E.
coli وK.
pneumonia و Aspergillus niger و Candida albicans و.Staphylococcus aureus نقي الكليبوسين جزئيا عن بروتينه المناعي بكروماتوغرافيا التبادل الأيوني كليا بكروماتوغرافيا الترشيح الهلامي و تأكدت نقاوته بواساطة الترحيل الهلامي وتمت SDS-PAGE بظهور حزمة بروتينية واحدة له.
كانت بكتريا Staphylococcus aureus الأكثر تثبيطا بينما K.pneumonia (K72) أقلها تثبيطا.
Abstract EN
-Klebsiella pneumonia (K64) isolated from urine was induced by mutagenic agent (Mitomycim C) to produce Klebocin.
The Klebocin was extracted and ultra filtrated then purified by Ion-exchange chromatography.
Klebocin was obtained by Gel-filtration chromatography ; the purified protein was applied to SDS-PAGE gel electrophoresis.
The antimicrobial activity was tested against different microorganisms that included: Escherichia coli, Klebsiella pneumonia, Staphylococcus aureus, Candida albicans and Aspergillus niger.
The Klebocin partially purified from its immunity protein by ion exchange chromatography and completely purified by gel filtration chromatography.
The purity was confirmed from one band obtained from electrophoresis.
Staphylococcus aureus was very sensitive while (K72) K.pneumonia revealed a minimum inhibition zone.
