A reverse micelle approach was developed to prepare liposome-encapsulated hemoglobin (LEH) as a red blood cell (RBG) substitute.
The new approach resulted in higher encapsulated hemoglobin (Hb) concentration than conventional approach (1-7).
An encapsulated 1 lb concentrations of about 20 gm% (gm/ 100 ml solution) were obtained which were at a level close to that of the precursor Hb concentration.
Two types of phosphatidylcholine were used with different processing parameters.
These parameters involved lipid weight, lipid/Hb loading, solvent type, buffering systems, and size reduction processes.
Quality parameters involved encapsulation efficiency, encapsulated Hb concentration, oxygen carrying capacity, and encapsulated Hb quality with respect to met-hemoglobin formation.
The investigated approach indicates a promising method to produce an excellent LEH quality.
