Background: A hallmark of arthritic conditions is the accumulation of leucocytes within the synovial fluid (SF) via the membrane.
Among those, cells of the monocytes/macrophages (M?) lineage play an important role in the pathogenesis of inflammatory joint diseases.
Objective: - to determine the presence of vascular cell adhesion molecule (VCAM), CD44 and CD15 on monocytes/ macrophage from peripheral blood (PB) and synovial fluid in patients with rheumatoid arthritis (RA), reactive arthritis (ReA) and osteoarthritis (OA) in order to improve our understanding of possible mechanisms underlying the inflammatory process.
Methods: - 45 patients were recruited for the study, they were equally divided into three groups according to the diagnosis of RA, ReA and OA.
They all had their PB and SF sampled for CRP, RF, CBC, ESR and SF-WBCs count.
Peripheral blood monocytes were separated and also, from SF and then stained with monoclonal antibodies for CD44, CD15 and VCAM.
Results: - In the RA group, CD44 was positive in 11 cases, CD15 positive in 10 cases and VCAM positive in 3 cases.
In the ReA group, CD44 was positive in 8 cases, CD15 positive in 10 cases and VCAM positive in 4 cases.
For OA group, CD44 was positive in 5 cases, CD15 positive in 7 cases and VCAM positive in 2 cases.
Conclusion:-It in concluded that CD44, CD15 and VCAM can not be used as a biological marker neither for blood nor from synovial fluid.
