تعرف بروتينات الصدمة الحرارية بأنها مواد يتم تكوينها داخل الخلية فسيولوجيا و كذلك تحت تأثير كثير من المؤثرات المهيجة للخلايا و هي تلعب دورا هاما في كثير من وظائف الخلية الهامة. إن هذه البروتينات قد تكون أحد أسباب أمراض المناعة الذاتية خاصة إذا كان هناك أحد العوامل التي تؤدي إلى زيادة تكوينها أو تغير موضعها داخل الخلية. إن واحدا من هذه البروتينات هو البروتين ذو الوزن الجزيئي – 90 كيلو دالتون و الذي قد يكون له دور في حدوث مرض الذئبة الحمراء. و الغرض من هذا البحث هو توضيح ما إذا كان بروتين الصدمة الحرارية – 90 يتم تكوينه بصورة زائدة في مرض الذئبة الحمراء و كذلك توضيح علاقته بنشاط المرض، و قد أجريت هذه الدراسة على 47 سيدة يتراوح عمرهن بين 11 – 47 عاما و قد تم تقسيمهن إلى مجموعتين : الأولى تشمل 34 مريض بالذئبة الحمراء و الثانية تشمل 13 سيدة سليمة و تمثل المجموعة الضابطة، و تم إجراء الاختبار الخاص بالكشف عن مستوى البروتين ذو الوزن الجزيئي – 90 داخل الخلايا الليمفاوية باستخدام طريقة Western Blotting. و لقد وجد أن هذا البروتين يتم تكوينه بصورة زائدة في 29.4% من الحالات التي تم دراستها بالمقارنة بالمجموعة الضابطة، كما وجد أن هناك علاقة بين تكوين هذا البروتين بصورة زائدة و نشاط المرض. و من هذا يتضح أن زيادة تكوين هذا البروتين قد تلعب دورا في إحداث مرض الذئبة الحمراء، لذا ننصح بعمل دراسات أخرى في هذا المجال لتحديد هذا الدور.
Abstract EN
Objective: Heat shock protein (HSP) is a group of constitutive proteins with important physiological functions whose synthesis is enhanced by elevated temperature and other stressful conditions.
Previous data suggested that HSP 90 might be involved in the etiology of autoimmune diseases.
The aim of this study was to assess the state of HSP 90 expression in systemic lupus erythematosus (SLE) and to clarify the relation between HSP 90 expression and disease process and activity.
Material & Methods: This study was performed on 34 SLE female patients and 13 control subjects of matching age and sex.
HSP 90 in peripheral blood mononuclear cells (PBMC) was detected using the Western blotting technique.
Results: HSP 90 was overexpressed in 29.4% (10 out of 34) of SLE patients and in none of the control group.
The HSP 90 overexpression was significantly associated with some parameters of disease activity.
Conclusion: Evidence suggests that HSP 90 may play a potential role in etiopathogenesis of SLE.
It provides a potential therapeutic target for this disease.
