English Abstract

In this study, new ethyl 7-( alkynyl)-1-cyclopropyl-6-fluoro-8-nitro-4-oxo-1,4-dihydro quinoline-3-carboxylates (44a-c) were prepared.

These derivatives were prepared through sonogashira cross-coupling reaction between ethyl 7-chloro-1-cyclopropyl-6-fluoro-8-nitro-4-oxo-1,4-dihydroquinoline -3-carboxylate (42) and some acetylene derivatives (43a-c) using Pd(ph3P)Cl2 , CuI and K2CO3 in DMF as a catalyste at controlled temperature (75-90 °C) The new compounds (44a-c) were obtained in moderate yields (42–55%).

Compounds (44a-c) were hydrolyzed to their carboxylic acid derivatives using HCl / EtOH under reflux temperature to give (45a-c) in high yields (64 – 72%).

Compounds (44a-c) and (45a-c) were characterized using different spectroscopic techniques such as: 1H-and 13C-NMR, ESI-Mass, FTIR spectroscopy and elemental analysis (C, H, N).

Antibacterial activity for compounds (45a-c) were tested using disk diffusion method, against Gram-positive and Gram-negative bacterial strain.

The study showed that compounds (45a-c) have antibacterial activity against only Gram positive bacterial strains
