الخلفية : خلال السنوات القليلة الماضية كان هناك نقاش واسع حول التبادلية و الفعالية بين المنتجات التي تحتوي على غليبينكلاميد المنشرة في السوق.
الأهداف : هدفت هذه الدراسة إلى مقارنة فعالية هذا المنتج "غير ميكرونيزيد" في الناتج المنشئ للأقراص غليبينكلاميد "من ميكرونيزيد" السلفونيل يوريا.
الطرق : تلقى 12 متطوعا جرعة من mg5 من غليبينكلاميد (من الاختبار و المنتجات القياسية) في ظل ظروف الصيام في دورتين منفصلتين باستخدام الطريقة العشوائية المتبادلة.
تم رصد مستوى السكر في الدم للمتطوعين لتجنب حدوث نقص السكر في الدم.
تم جمع عينات البلازما على مدى 24 ساعة و تحليلها باستخدام HPLC.
النتائج : كان التركيز الأقصى للـ CMAX للاختبار و المنتجات المرجعية 2.508 ± 0.104 و 3.526 ± 0.118 (ميكروغرام / مل) على التوالي و المنطقة تحت المنحنى AUC0-∞ كانت 3.511 ± 0.153، 4.572 ± 0.202 (μg.h / مل) لهذه المنتجات على التوالي.
مع وجود فرق من حوالي 24 ٪ بين منتجات الاختبار و القياس في AUC.
الاستنتاجات : و تشير النتائج إلى أن منتج الاختبار ليس مكافئا بيولوجيا لمنتج القياس.
الفرق في الصياغة بين المنتج ميكرونيزيد و المنتجات غير ميكرونيزيد من أقراص غليبينكلاميد له تأثير على النتائج السريرية.
Abstract EN
-Background : During the last few years there was wide debate about the interchangeability and effectiveness between circulated products containing Glibenclamide in the market.
Objectives : This study aimed to compare the effectiveness of this product !non-micronized" to the originator#s product of Glibenclamide tablets !of micronized" sulfonylurea.
Methods : 12 volunteers received a dose of 5mg of Glibenclamide (from test and standard products) under fasting conditions in two separate sessions using randomized crossover design.
Blood glucose level for the volunteers was monitored to avoid the development of hypoglycemia.
Plasma samples were collected over 24 hours and analyzed using HPLC.
