تم تحقيق إنتاج الكالس، مرحلة التطور الجنيني الجسدي على الكالوس و إعادة تخليق النبات من الحويصلات العصيرية في الليمون المكسيكي.
أولا، تم استخدام ثلاثة أنواع من الاوساط الغذائية، اولها بيئة موراشيج و سكوج (MS) and Skoog Murashige) و الثانية موراشيج و تاكر (MT) و غروسير (H + H) و مستويات مختلفة من 4٬2-Dicholorophenoxy (D-4,2) لدراسة تأثيرها على الإنتاج و انتشار الكالس الجنيني من الحويصلات العصيرية.
ثانيا، لبدء الأجنة الجسدية من الكالس، فقد جرى التحقيق من خمسة أنواع من الأوساط الغذائية المختلفة و عشرين مزيج من معاملات منظمات النمو.
و أظهرت النتائج أنبيئة موراشيج و سكوج MS المزودةب5.0 ملغ / لتر D-4,2 إنتجت أقصى قدر من الكالس الجنيني.
في بيئي MT وH H +، تم الحصول على أقصى قدر من الكالس مع 5.0 و5.lملغم / لتر من D-4,2، على التوالي.
تشكلت الأجنة الجسدية فقط عن طريق استخدام وسط MT الذي يحتوي على 500-700 ملغم / لتر من مستخلص الشعير.
الأجنة الجسدية نبتت و أكملت نموها على بيئة موراشيج و سكوج MS مع 0.l ملغ / لتر حمض الجبريليك الحصول على أقصى طول ساق و عدد الأوراق عندما أضيف 0.l ملغ / لتر من حامض الجبريلين GA3 إلى بيئة موراشيج و سكوج MS.
تم نقل شتلات في قدور تحتوي على نسبة l : 2 : 2 بنسبة الحجم من خليط التربة (الرمل و البيتموس و البيرلايت، على التوالي)، و كان معدل البقاء على قيد الحياة للشتلات بنسبة l00 ٪.
Abstract EN
Callus production, somatic embryogenesis and plant regeneration were achieved from juice vesicles of Mexican lime.
First, three types of culture media, Murashige and Skoog (MS), Murashige and Tucker (MT) and Grosser and Gmitter (H+H) and different levels of 2,4-Dicholorophenoxy acetic acid (2,4-D) were used to study their effects on production and proliferation of embryogenic calli from juice vesicles.
Second, for initiation of somatic embryos from embryogenic calli, five different media and twenty combinations of growth regulator treatments were investigated.
Results showed that MS medium supplemented with 0.5 mg/l 2,4-D produced maximum embryogenic calli.
In MT and H+H media, maximum calli were obtained with 0.5 and 1.5 mg/l 2,4-D, respectively.
Somatic embryos were formed only by the use of MT medium containing 500 to 700 mg/l malt extract.
Embryos germinated and completed their growth on MS medium supplemented with 1.0 mg/l Gibberlic acid (GA3) and 0.02 mg/l α-Naphthalene acetic acid (NAA).
Maximum shoot length and leaf number were observed when 1.0 mg/l GA3 was added to the MS medium.
Plantlets were transferred into pots containing of 1:2:2 volume ratio of soil mixture (sand, peat moss and perlite, respectively) and the rate of survival was 100%.
