English Abstract
Human hydatid cysts were surgically removed from 30 patients who underwent surgery for hepatic, pulmonary, spleen, heart, and peritoneal cavity cystic echinococcosis (CE) at hospitals from various provinces of Iraq, between March 2012 and January 2013.
According to questionnaire sheet that used for demographic, epidemiologic, and clinical data for each patients, the result showed the incidence of CE was higher in females than in males and most infections 33.33% were at the age group (21-30) year.
The proportion of infection in rural areas were higher than urban, with most affected organ the liver 50%, cysts integrity characterized by high percentage (90%) of intact cyst.
Two components of hydatid cysts (germinal layer and protoscoleces) were used for DNA extraction by four different methods (mechanical grinder, boiling, freezing-thawing, and crushing by sonicator).
The amount of released DNA from protoscoleces was higher than germinal layer and mechanical grinding obtained best results for this process.
Purification of mtDNA was done by three procedures (manual extraction, promega kit, and genaid kit) and promega kit was selected for its advantages in comparison to the other two procedures.
The mitochondrial COX1 (cytochrome c oxidase subunit 1) and NADH dehydrogenase subunit 1 (ND1) genes were used as targets for polymerase chain reaction (PCR) amplification, all of 30 hydatid cysts yielded amplification products.
For CO1 primers produced 450 bp PCR product while for NADH1 primers produced 800 bp.
IV The PCR products were purified and partial sequences were generated.
The sequences obtained were found to align with corresponding region for CO1 and ND1 genes in the Gene Bank nucleotide database confirming the human cysts to contain genotype of sheep strain (G1) in Iraq.
The sequences were further aligned with the corresponding regions of CO1 and ND1 genes of known genotypes from other countries to compare the degree of matching especially with other countries and the results showed that the highest match (26.66%) of CO1 was with Mongolia.
While, for ND1 the highest match (70%) with Argentina.
