English Abstract

Immunoradiometric assay technique was used to measure the level of serum tumor marker Alpha- fetoprotein (AFP) in: 25 healthy women, 79 patients with different histological types of benign and malignant ovarian tumors respectively.

Upon comparison of the results, the AFP level in sera of patients with malignant ovarian tumors was found to be significantly higher (73.71 ± 104.89 IU.ml-1) (P < 0.05) than their corresponding values in sera of healthy (7.98 ± 2.79 IU.ml-1) and that in patients with benign tumors (10.38 ± 8.36 IU.ml-1).

The mean value of AFP tumor marker in relation to the different histological types of tumors, was found to be significantly higher (P < 0.05) in malignant germ cell ovarian tumors(171.27 ± 130.69 IU.ml-1), than in other types of benign and malignant tumors.

Immunoradiometric assay (IRMA) was modified and used to characterize the binding of 125I-anti AFP antibody to AFP of ovarian tumor tissues.

The data obtained revealed an increment of AFP in the supernatant fraction in comparison to the pellet part.

Also a higher incidence of AFP in malignant than those in benign ovarian tumor tissues.

The characteristics of the binding of 125I-anti AFP antibody with its AFP antigen of benign germ cell (OI) and malignant germ cell (OII) and of malignant serous (OIII) ovarian tumor tissues was investigated.

Different factors affecting this binding were extensively studied such as pH, time, temperature, concentration of antibody, concentration of antigen and different concentrations of salts.

The antigen AFP obtained from malignant germ cell ovarian tumor tissue homogenate (OIII) was isolated by gel filtration chromatography using sepharose CL-6B (0.96 × 29 cm) where three immunoreactive forms of AFP were separated peak I (PI), peak II and peak III (PIII) with molecular weight of 592, 190 and 77 KDa respectively.

The binding characteristics of the isolated AFP third form (PIII) with 125I-anti AFP antibody were investigated.

Kinetic parameters of the binding of 125I-anti AFP antibody with AFP of the homogenates obtained from benign germ cell (OI), malignant germ cell tumors (OII), malignant serous tumors (OIII) , and isolated AFP (PIII), at different temperatures V were determined.

The results indicated that the binding reaction was time and temperature dependent process.

The order of the binding reactions were tested using different rate equations.

In addition, the kinetic parameters ka, kd, k+1, k-1 , Hill coefficient, t½ of association and dissociation were determined at 4,25, 37 and 45 ˚C.

The thermodynamic of the binding of 125I-anti AFP antibody with AFP of benign germ cell (OI), malignant germ cell (OII), malignant serous ovarian tumor homogenates (OIII) and with isolated AFP of malignant germ cell ovarian tumor homogenates (PIII) were studied, using Van't Hoff and Arrhenius equations.

The thermodynamic parameters of the standard state ( ΔH˚, ΔG˚, ΔS˚) and the transition state (Ea, ΔH*, ΔG*, ΔS*) were also determined.

Spectroscopic studies in the U.V region, were carried out on human AFP and the complex formed (125I-anti AFP antibody /AFP) of isolated AFP of malignant germ cell ovarian tumors.

Different factors affecting the absorption band such as pH, polarity and denaturation agents were extensively studied.

The heat stability and spectrophotometer pH titration of the complex were also include in this work.
